Together, these two proteins synergistically activate the TAK1/JNK1 signal transduction pathway, which acts to protect against the interleukin-1b converting enzyme of Fas-induced apotosis. This protein was independently isolated and characterized as polyploidy-associated protein kinase (PAPK), a member of the Ste20/germinal center kinase family that modulates cytoskeletal organization and cell survival, suggesting that its protective role may be a function of its kinase activity. Although two mRNA sequences have been isolated, only the shorter isoform has been observed.

Lano is a member of the LAP (leucine-rich repeats and PDZ) family of proteins that also includes Densin-180, Erbin, and hScribble. The LAP proteins generally contain multiple leucine-rich repeat (LRR) domains which serve to target them to the basolateral membrane of epithelial cells. Lano is unique in that it alone does not possess one or more PDZ (PSD95/DLG/ZO-1) domains as do the other members of the LAP family. However, it can bind to the PDZ domain of Erbin in addition to those of membrane-associated and guanylate kinase (MAGUK) proteins which regulate adhesion and plasticity at cell junctions. It has been suggested that it is through these interaction that these LAP proteins participate in the maintenance of proper embryonic development and integrity of epithelial tissues.

PLEKHM1 is a member of the M family of Pleckstrin homolog domain-containing proteins, a group of proteins containing a RUN domain, two pleckstrin homology domains, and a cysteine-rich domain. It was identified through segregation analysis as a cause of osteopetrosis in humans. PLEKHM1 co-localizes with Rab7 to late endosomal/lysosomal vesicles in HEK293 and osteoclast-like cells, with this co-localization dependent on the prenylation of Rab7. Monocytes from a patient homozygous for a mutated form of PLEKHM1differentiated into osteoclasts normally, but failed to form ruffled borders and showed little evidence of bone resorbtion when cultured on dentine discs. Another mutation of PLEKHM1 impaired vesicular acidification and increased TRACP secretion in osteoclasts, suggesting that PLEKHM1 has critical roles in endosomal maturation and may be important in osteoclast-osteoblast cross-talk.
