Multiple isoforms of CD45R are distributed throughout the immune system according to cell type. These isoforms arise because of alternative splicing of exons 4, 5, and 6. The corresponding protein domains are characterized by the binding of monoclonal antibodies specific for CD45RA (exon 4), CD45RB (exon 5), CD45RC (exon 6) and CD45RO (exons 4 to 6 spliced out). The variation in these isoforms is localized to the extracellular domain of CD45R, while the intracellular domain is conserved. CD45R functions as a phosphor-tyrosine phosphatase. Antibody to CD45 is useful in differential diagnosis of lymphoid tumors from non-hematopoietic undifferentiated neoplasms.

This mAb reacts with a wide variety of simple epithelia. It does not react with stratified squamous epithelia. It reacts with epithelial tumors of the gastrointestinal tract, lung, breast, pancreas, ovary, and thyroid. Cytokeratin 18, which belongs to the type A (acidic) subfamily of low molecular weight keratins, exists in combination with cytokeratin 8. It is reported that tissues from gastrointestinal tract are positive for both cytokeratin 8 and 18 but do not contain cytokeratin 14. Tissues from gastrointestinal tract, respiratory tract and urogenital tract, as well as endocrine and exocrine tissues and mesothelial cells are positive for cytokeratin 18.

Recognizes 78kDa moesin protein. Moesin, a member of the talin-4.1 superfamily, is a linking protein of the sub-membranous actin cytoskeleton. It is expressed in variable amounts in cells of different phenotypes such as macrophages, lymphocytes, fibroblastic, endothelial, epithelial, and neuronal cell lines but not in blood cells. The ERM proteins, ezrin, radixin, and moesin are involved in a variety of cellular functions, such as cell adhesion, migration, and the organization of cell surface structures, and are highly homologous, both in protein sequence and in functional activity, with merlin/schwannomin, a neurofibromatosis-2-associated tumor-suppressor protein. Cell lines of epithelial and mesothelial origin contain both moesin and radixin whereas cells of endothelial and lymphoid origin express moesin.

Early Mitotic Inhibitor-1 regulates mitosis by inhibiting the anaphase promoting complex/cyclosome (APC). It is a conserved F box protein containing a zinc-binding region essential for APC inhibition. The protein functions to promote cyclin A accumulation and S phase entry in somatic cells by inhibiting the APC complex. At the G1-S transition, EMI-1 is transcriptionally induced by the E2F transcription factor. Overexpression accelerates S-phase entry and can override a G1 block caused by overexpression of Cdh1 or the E2F-inhibitor p105 retinoblastoma protein (pRb). Depleting cells of EMI-1 through RNA interference prevents accumulation of cyclin A and inhibits S phase entry.

Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Most pituitary adenomas and prolactinomas readily express chromogranin.

This antibody recognizes a 180-185kDa protein, identified as isoform of leukocyte common antigen (CD45) called CD45RO (4th Leucocyte Typing Workshop: Code No. N31). The epitope recognized by this antibody is sensitive to neuraminidase digestion. CD45RO is found on mature activated T-cells, most thymocytes, and a sub-population of resting T-cells within both CD4 and CD8 subsets. The CD45RO antibody shows no reactivity with normal B or natural killer cells, but reacts with granulocytes and monocytes. Reportedly, antibody to CD45RO is useful to identify T-cell lymphomas and leukemias. It rarely stains NK cells or B-cell lymphomas.

This antibody recognizes CK1, CK5, CK10 and CK14. In normal epithelia, it stains stratified epithelia, myoepithelial cells and basal cells in the prostate gland and bronchi. This mAb shows no reactivity with hepatocytes, pancreatic acinar cells, proximal renal tubules, or endometrial glands; there is no reactivity with cells derived from simple epithelia. Mesenchymal tumors, lymphomas, melanomas, neural tumors, and neuroendocrine tumors are negative with this antibody. It stains myoepithelial cells and has been shown to be useful in distinguishing prostate adenocarcinoma from benign prostate. This antibody has also been useful in separating benign from malignant intraductal breast proliferations.

This antibody recognizes proteins of 80-200kDa, identified as different members of CEA family. CEA is synthesized during development in the fetal gut and is re-expressed in increased amounts in intestinal carcinomas and several other tumors. This mAb reacts with nonspecific cross-reacting antigen (NCA). It shows no reaction with a variety of normal tissues and is suitable for staining of formalin/paraffin tissues. CEA is not found in benign glands, stroma, or malignant prostatic cells. Antibody to CEA is useful in detecting early foci of gastric carcinoma and in distinguishing pulmonary adenocarcinomas (60-70% are CEA+) from pleural mesotheliomas (rarely or weakly CEA+). Anti-CEA positivity is seen in adenocarcinomas from the lung, colon, stomach, esophagus, pancreas, gallbadder, urachus, salivary gland, ovary, and endocervix.

It recognizes a protein of HMW, identified as mucin 3 glycoprotein (MUC3). This mAb shows no cross-reaction with human milk fat globule membranes, MUC1, or MUC2. The Mucins are a family of highly glycosylated, secreted proteins with a basic structure consisting of a variable number of tandem repeats (VNTRs) encoded by 60 base pairs (Mucin 1), 69 base pairs (Mucin 2) and 51 base pairs (Mucin 3). The tetraspanin CD63 is a lysosomal membrane glycoprotein that translocates to the plasma membrane after platelet activation. CD63 is expressed on activated platelets, monocytes and macrophages, and is weakly expressed on granulocytes, T cell and B cells. It is located on the basophilic granule membranes and on the plasma membranes of lymphocytes and granulocytes. CD63 is a member of the TM4 superfamily of leukocyte glycoproteins that includes CD9, CD37 and CD53, which contain four transmembrane regions. It binds to MHC class 1 and through its association with protein tyrosine kinase p56lck plays a role in Tcell development and activation of mature Tcells. For mature Tcells, CD8 and 4 are mutually exclusive, so antibody to CD8 is generally used in conjunction with antibody to CD4. CD8a antibody is a useful marker for distinguishing helper/inducer T-lymphocytes, and most peripheral Tcell lymphomas are CD4+/CD8-. This antibody recognizes a protein doublet of 20-22kDa, identified as MART-1 (Melanoma Antigen Recognized by T cells 1) or Melan-A. MART-1 is a melanocyte differentiation antigen recognized by autologous cytotoxic T lymphocytes. There are seven other melanoma associated antigens recognized by autologous cytotoxic T cells: MAGE-1, MAGE-3, tyrosinase, gp100, gp75, BAGE-1, and GAGE-1. Subcellular fractionation shows that MART-1 is present in melanosomes and endoplasmic reticulum. This MART-1 antibody labels melanomas and other tumors showing melanocytic differentiation. It is also a useful positive-marker for angiomyolipomas. The antibody does not stain tumor cells of epithelial, lymphoid, glial, or mesenchymal origin.
