yDcpS

yDcpS

Supplier: New England Biolabs (NEB)

yDcpS is the scavenging decapping enzyme from S. cerevisiae that hydrolyzes the phosphodiester bond between the gamma and beta phosphates of m7G-capped mRNA, producing diphosphorylated 5 ends and m7GMP. yDcpS is capable of decapping mRNAs of various lengths, decaps Cap0 and Cap1 with equal efficiency, and the diphosphorylated 5 end it produces is suitable for recapping with Vaccinia capping enzyme.

JTE-607 >98% mRNA processing inhibitor

Supplier: G-Biosciences

Inhibits inflammatory cytokine production in human peripheral blood mononuclear cells (PBMC’s) without causing immunosuppression: IC50’s = 11 nM (TNF-α), 5.9 nM (IL-1β), 8.8 nM (IL-6), 7.3 nM (IL-8), and 9.1 nM (IL-10).1 It displayed efficacy in a mouse model of septic shock.2 JTE-607 also showed inhibitory activity against acute myelogenous leukemia cell lines.3,4 Recently, the mechanism of action of JTE-607 (a pro-drug, with the active species being the free acid) has been found to be inhibition of pre-messenger RNA endonuclease Cleavage and Polyadenylation Specificity Factor 3 (CPSF3).5,6 This prevents release of newly synthesized mRNA’s resulting in read-through transcription and the formation of DNA-RNA hybrid R-loop structures. Transcripts down-regulated by JTE-607 were related to DNA damage-based phenotype.

RECTAS >98% pre-mRNA splicing modulator

Supplier: G-Biosciences

RECTAS a pre-mRNA splice modulator (Rectifier of Aberrant Splicing) was discovered in a small molecule screen for compounds that rectify aberrant IKBKAP splicing in cells from patients with familial dysautonomia. RECTAS (2 µM) increased the expression of IKAP and restored tRNA modifications1,2. It directly interacts with CDC-like kinases enhancing SRSF6 phosphorylation2. A combination treatment with RECTAS and phenylbutyric acid was shown to restore DJ-1 protein and mitochondrial dysfunction in Parkinson’s disease models.

Anti-LIN28 Rabbit Polyclonal Antibody

Anti-LIN28 Rabbit Polyclonal Antibody

Supplier: Prosci

LIN28 acts as a 'translational enhancer', driving specific mRNAs to polysomes and thus increasing the efficiency of protein synthesis. The association of LIN28 with the translational machinery and target mRNAs results in an increased number of initiation events per molecule of mRNA and, indirectly, in stabilizing the mRNAs. IT binds IGF2 mRNA, MYOD1 mRNA, ARBP/36B4 ribosomal protein mRNA and its own mRNA. LIN28 is Essential for skeletal muscle differentiation program through the translational up-regulation of IGF2 expression.

Anti-DCP1B Rabbit Polyclonal Antibody

Anti-DCP1B Rabbit Polyclonal Antibody

Supplier: Prosci

DCP1B may play a role in the degradation of mRNAs, both in normal mRNA turnover and in nonsense-mediated mRNA decay. It may remove the 7-methyl guanine cap structure from mRNA molecules, yielding a 5'-phosphorylated mRNA fragment and 7m-GDP.DCP1B is a core component of the mRNA decapping complex, a key factor in the regulation of mRNA decay (Lykke-Andersen, 2002 [PubMed 12417715]).

Anti-DCP2 Rabbit Polyclonal Antibody

Supplier: Rockland Immunochemical

The removal, or decapping, of eukaryotic mRNA is an important step in the degradation of mRNA. Decapping protein 2 (DCP2) is the major mRNA decapping enzyme in cells. It is a member of the Nudix hydrolases superfamily of proteins that predominantly catalyze the hydrolysis of small nucleoside diphosphate substrates linked to another moiety. DCP2 is widely expressed in multiple tissues at varying levels, with highest expression seen in testis and brain.

Anti-DCP2 Rabbit Polyclonal Antibody

Anti-DCP2 Rabbit Polyclonal Antibody

Supplier: Prosci

DCP2 is necessary for the degradation of mRNAs, both in normal mRNA turnover and in nonsense-mediated mRNA decay. DCP2 removes the 7-methyl guanine cap structure from mRNA molecules, yielding a 5'-phosphorylated mRNA fragment and 7m-GDP. It has higher activity towards mRNAs that lack a poly (A) tail, but has no activity towards a cap structure lacking a RNA moiety.DCP2 is a key component of an mRNA-decapping complex required for removal of the 5-prime cap from mRNA prior to its degradation from the 5-prime end (Fenger-Gron et al., 2005 [PubMed 16364915]).

Anti-THOC2 Rabbit Polyclonal Antibody (HRP (Horseradish Peroxidase))

Supplier: Bioss

Required for efficient export of polyadenylated RNA and spliced mRNA. Acts as component of the THO subcomplex of the TREX complex which is thought to couple mRNA transcription, processing and nuclear export, and which specifically associates with spliced mRNA and not with unspliced pre-mRNA. TREX is recruited to spliced mRNAs by a transcription-independent mechanism, binds to mRNA upstream of the exon-junction complex (EJC) and is recruited in a splicing- and cap-dependent manner to a region near the 5' end of the mRNA where it functions in mRNA export to the cytoplasm via the TAP/NFX1 pathway. The TREX complex is essential for the export of Kaposi's sarcoma-associated herpesvirus (KSHV) intronless mRNAs and infectious virus production. THOC2 (and probably the THO complex) is involved in releasing mRNA from nuclear speckle domains. Required for NXF1 localization to the nuclear rim.
