Murine CD38, a type II transmembrane glycoprotein, is a bifunctional ectoenzyme capable of catabolizing nicotinamide adenine dinucleotide (NAD+) to cyclic ADP-ribose (cADPR), and then hydrolyzing cADPR to adenosine diphosphoribose (ADPR). It is expressed at high levels on the surface of peripheral B-lineage cells and at low density on germinal center B cells from unimmunized mice. It has also been reported to be expressed at moderate levels on NK cells, a proportion of peripheral T cells, and a subpopulation of thymocytes which are mostly TCRαβ+, CD4-, CD8-. Murine CD38 is also expressed by all Mac-1+ macrophages in the peritoneal cavities of unimmunized mice but not by unstimulated bone-marrow-derived macrophages. Monoclonal antibodies to CD38 have been shown to induce B and T cell proliferation, protect B cells from apoptosis, and inhibit B lymphopoiesis.

CD69, also known as very early activation (VEA) antigen, is a disulfide-linked transmembrane homodimer whose differentially glycosylated subunits range from 35-39 kDa. It is a C-type lectin most closely related to the NKR-P1 and Ly-49 NK cell-activation molecules. CD69 is widely expressed on hematopoietic cells, including lymphocytes, neutrophils and eosinophils. Although not detectable on resting lymphocytes, its expression is rapidly upregulated upon activation of T, B and NK cells, and neutrophils. Constitutive expression of CD69 on subsets of thymocytes suggests that it may be involved in regulation of developmental events in addition to its role in activation of a variety of hematopoietic cells. The “classical” MHC Class I molecules are histocompatibility antigens encoded by the H-2 gene complex and consist of heterodimers of highly polymorphic α chains noncovalently associated with the invariant β2-microglobulin. These antigens are expressed on most nucleated cells but expression varies on different cell types. MHC Class I molecules present endogenously synthesized peptides to CD8+ T lymphocytes which are usually cytotoxic T cells. MHC Class I antigens expressed on thymic epithelial cells regulate the positive and negative selection of CD8+ T cells during T cell ontogeny. The monoclonal antibody 28-14-8 binds to the α3 domain of H-2Db in the presence or absence of β2 microglobulin. It cross reacts with the α3 domain of H-2Ld, but not Kd or Dd, and with H-2Dq and/or Lq. The antibody has been shown to block H-2Ld-specific and H-2Ld-restricted antigen-specific lysis of target cells by cytotoxic T lymphocytes but it does not block recognition of H-2Ld-positive target cells by Ly-6G2-positive NK cells.

CD106, also known as INCAM-110, is a 110 kDa vascular adhesion cell adhesion molecule-1 (VCAM-1) that is member of the immunoglobulin superfamily. CD106 is expressed predominantly on cytokine-activated vascular endothelium but has also been identified on interfollicular dendritic cells, some macrophages, and bone marrow stromal cells. Endothelial CD106 binds the integrins α4β1 (CD49d/CD29, VLA-4) and α4β7 and contributes to extravasation of lymphocytes, monocytes, basophils, and eosinophils (but not neutrophils) from blood vessels, particularly at sites of inflammation. Unlike the β2 integrins, the CD106-VLA-4 interaction can mediate both the initial tethering and rolling of lymphocytes on endothelium as well as their subsequent arrest and firm adhesion. CD106 expressed on non-vascular tissues has been implicated in the interaction of hematopoietic progenitors with bone marrow stromal cells, B cell binding to follicular dendritic cells, costimulation of T cells, and embryonic development. The monoclonal antibody 1.G11B1 inhibits in vitro binding of lymphocytes and monocytes to VCAM-1 on stimulated endothelium.

CD30 is a type I transmembrane protein and a member of the tumor necrosis factor receptor superfamily of proteins. Murine CD30 is expressed predominantly in the thymus and it is inducible in mouse splenocytes stimulated with pokeweed mitogen or concanavalin A. In anti-CD3ε-activated spleen cells, CD30 is expressed primarily on the surface of CD8+ T cells with peak expression on days 4 and 5. Stimulation of CD30+ CTL lines with plate-bound anti-CD30 directly signals IL-5 but not IFN-γ production. While these studies demonstrate that CD30 directs cytokine secretion and suggest that CD30 may play a pivotal role in the pattern of cytokine production by T cells, the precise roles of CD30 and its ligand (CD153) in T-cell development have not been clearly defined.
