Apoptosis plays a major role in normal organism development, tissue homeostasis, and removal of damaged cells. Disruption of this process has been implicated in a variety of diseases such as cancer. The Bcl-2 family of proteins is comprised of critical regulators of apoptosis that can be divided into two classes: those that inhibit apoptosis and those that promote cell death. Bid, a pro-apoptotic Bcl-2 family member, is cleaved by caspase-8 in response to apoptotic signals, exposing the Bcl-2 homology 3 (BH3) domain which is normally buried in the full-length protein. The cleaved complex is myris-toylated and translocated to the mitochondrial membrane where it may induce mitochondrial Bax and Bak to oligomerize.

The proper expression of gene products in eukaryotic cells relies on efficient transport of mRNA molecules out of the nucleus. Gle1 is an essential mRNA export factor in both human and yeast cells. It associates with the nuclear pore complex (NPC) through hCG1 and NUP155 in mammalian cells and in conjunction with inositol hexakisphosphate (IP6), stimulates Dbp5, a member of the DEAD-box helicase family, triggering mRNP remodeling and facilitating RNA export from the nucleus. HEPACAM2 (Hepatocyte cell adhesion molecule 2), a type I N-linked transmembrane glycoprotein, belongs to the immunoglobulin superfamily. The exact function of HEPACAM2 is currently unknown, but the related protein HEPACAM forms cishomodimers on the cell surface to regulate the cell adhesion and may inhibit cell growth through suppression of cell proliferation. HEPACAM and HEPACAM2 mRNA are differentially regulated in canine tumors, with HEPACAM2 mRNA showing increased levels in adenomas, but decreased levels in metastatic carcinomas compared to normal tissues, while HEPACAM protein levels decreased in adenomas. It is therefore likely that HEPACAM2 plays a different role than HEPACAM in the development and progression to tumors.

THEM2, also known as ACOT13, belongs to the hotdog-fold superfamily and possesses thioesterase activity, with greater activity observed with longer chain acyl-CoAs such as myristoyl- and palmitoyl-CoA. THEM2 is highly expressed in several tissues such as heart, kidney, liver and brain and has been shown to be co-localized with beta-tubulin on microtubules. THEM2 interacts with StarD, a protein that plays a key role in fatty acid metabolism, and the addition of StarD to THEM2 increased its catalytic activity, suggesting that THEM2 plays a significant role in the metabolism of fatty acids.
