N ethylmaleimide sensitive factor (NSF) is an ~83 kDa protein (predicted MW) which was initially isolated based on its ability to restore intercisternal Golgi transport to N ethylmaleimide treated membranes. NSF was subsequently discovered to be involved in a variety of membrane fusion events. NSF is an ATPase with two ATP binding domains that are essential for membrane fusion and homo oligomerization. In order to interact with target membranes, NSF requires another set of soluble proteins called Soluble NSF attachment proteins (SNAPs). NSF has been shown to play a prominent role in synaptic vesicle fusion. Together with synaptotagmin and a SNAP, NSF modulates the interaction of SNAP25, VAMP, and syntaxin, in an ATP dependent manner, to form a complex with a sedimentation coefficient of 20 S. This complex may represent an intermediate involved in synaptic vesicle docking and fusion.

Retention of resident soluble proteins in the lumen of the endoplasmic reticulum (ER) is achieved in both yeast and animal cells by their continual retrieval from the cis-Golgi, or a pre-Golgi compartment. Sorting of these proteins is dependent on a C-terminal tetrapeptide signal, usually lys-asp-glu-leu (KDEL) in animal cells, and his-asp-glu-leu (HDEL) in S. cerevisiae. This process is mediated by a receptor that recognizes, and binds the tetrapeptide-containing protein, and returns it to the ER. In yeast, the sorting receptor encoded by a single gene, ERD2, which is a seven-transmembrane protein. Unlike yeast, several human homologs of the ERD2 gene, constituting the KDEL receptor gene family, have been described. The protein encoded by this gene was the first member of the family to be identified, and it encodes a protein structurally and functionally similar to the yeast ERD2 gene product.

Nuclear factor kappa B (NF-kB) is a ubiquitous transcription factor and an essential mediator of gene expression during activation of immune and inflammatory responses. NF-kB mediates the expression of a great variety of genes in response to extracellular stimuli. NF-kB is associated with IkB proteins in the cell cytoplasm, which inhibit NF-kB activity. Phosphorylation of I-kB by IkB kinase (IKK) complex leads to degradation of I-kB and activation of NF-kB. The IKK complex contains IKKa,IKKb, and IKKg. A novel IKK related kinase was recently identified and designated TBK1 (TANK-binding kinase 1), NAK (NF-kB-activating kinase), and T2K. NAK/TBK1 activates IKKb through direct phosphorylation. NAK/TBK1is activated by growth factors and PMA and mediates IKK and NF-kB activation in response to growth factors. NAK/TBK1 functions upstream of NIK and the IKK complex. NAK/TBK1 is also critical in protecting embryonic liver from apoptosis.

CD40 (48 to 50 kDa) is a transmembrane glycoprotein mainly expressed on the surface of B cells and also expressed on monocytes, dendritic cells, and thymic epithelium. CD40 is a member of the tumor necrosis factor (TNF) receptor superfamily, which includes the low affinity nerve growth factor (NGF) receptor and CD95/Fas. CD40 is the receptor for CD40 ligand. CD40 ligand (CD40L, CD154, gp39, and TRAM) belongs to the TNF gene family and is expressed more widely than CD40 predominantly on activated CD4+ T cells. Following interaction with CD40 ligand, CD40 mediates a number of major immunoregulatory functions, central to the control of thymus dependent humoral immunity and may be critical in the development of cell mediated immune responses. Other biological actions include B cell homotypic adhesion, proliferation, immunoglobulin isotype switch, and secretion. Activation of CD40 has also been shown to inhibit the growth of certain B cell lymphomas and to induce the death of transformed cells of mesenchymal or epithelial origin.

Several novel death receptors including DR3, DR4, DR5, and DR6 were recently identified. Cell death signal is transduced by death domain containing adapter molecules through the interaction with death domain of these death receptors. A novel TNF-R1 interacting protein was recently identified and designated SODD for silencer of death domains. SODD associates with the death domain of TNF-R1 and prevents constitutive activation of TNF-R1 signaling. TNF treatment releases SODD and permits adapter molecules such as TRADD recruiting to the active TNF-R1 complex, which activates TNF signaling pathways. SODD also interacts with DR3. SODD is ubiquitously expressed in human tissues and cell lines.
