Tricine was first prepared by Good for use as a buffer for chloroplast reactions. It is structurally similar to Tris, but is much less inhibitory at high concentrations. For ATP assays using firefly luciferase, tricine buffer at 25 mM was found to be the best of ten common buffers tested.
Tricine is used as buffer component for separation of low molecular weight peptides. Tricine can be used in cryopreservation medium for the preservation of tissues and organs. Cryopreservation depends on the physical and chemical characteristics of the preservation medium used. The pH values and pK values for tricine/DMSO mixtures has been reported down to -20 °C. Tricine has been found to be an efficient scavenger of hydroxyl radicals in a study of radiation-induced membrane damage. Tricine is typically the buffer of choice in SDS-PAGE systems when separating proteins in the range of 1 to 100 kDa.
A buffer may be prepared by titrating with sodium hydroxide to the desired pH, using about a half-equivalent of NaOH.

Thermo Scientific Pierce DSP (Lomant's Reagent) is a water-insoluble, homo-bifunctional N-hydroxysuccimide ester (NHS ester) crosslinker that is thiol-cleavable, primary amine-reactive, and useful for many applications. DSP contains an amine-reactive NHS ester at each end of an 8-carbon spacer arm. NHS esters react with primary amines at pH 7–9 to form stable amide bonds and releasing N-hydroxy-succinimide. Proteins, including antibodies, generally have several primary amines in the side chain of lysine (K) residues and the N-terminus of each polypeptide that are available as targets for NHS ester crosslinking reagents. DSP is non-sulfonated and insoluble in water, so it must first be dissolved in an organic solvent and then added to the aqueous reaction mixture. Because DSP does not possess a charged group, it is lipophilic and membrane-permeable and so useful for intracellular and intramembrane conjugation. A sulfonated analog of DSP (DTTSP) is water soluble. DSS, the non-cleavable analog of the DSP crosslinker is also available for applications that require a stable spacer arm that cannot be cleaved in the presence of reducing agents.

Pyruvic acid is an intermediate in sugar metabolism and in enzymatic carbohydrate degradation (alcoholic fermentation) where it is converted to acetaldehyde and CO2 by carboxylase. In muscle, pyruvic acid (derived from glycogen) is reduced to lactic acid during exertion, which is reoxidized and partially retransformed to glycogen during rest. It improves coliform recovery when present in culture medium. It is involved in a metabolic regulatory pathway activated by mitochondrial oxidants. Pyruvate is involved in respiratory regulation in plants by interacting with alternative oxidase at a conserved cysteine residue. It may help prevent hydrogen peroxide mediated cell death.
Sodium pyruvate is utilized as a component in culture broth and media. Sodium pyruvate is used in Wallen fermentation medium to enhance the conversion of oleic acid to 10-ketostearic acid by Bacillus sphaericus. Sodium pyruvate has also been used to establish stably transfected human B cell lines.
Sodium Pyruvate has shown antioxidant properties and protective effects against oxygen radicals. Pyruvate is produced as part of glycolysis and is an intermediate in many metabolic pathways. It can be converted into acetyl CoA and enter the TCA Cycle.

The intermediate is unstable in aqueous solutions and so two-step conjugation procedures rely on N-hydroxysuccinimide (NHS) for stabilization. Failure to react with an amine will result in hydrolysis of the intermediate, regeneration of the carboxyl, and release of an N-substituted urea. A side reaction is the formation of an N-acylurea, which is usually restricted to carboxyls located in hydrophobic regions of proteins.

Cycloheximide is a glutarimide antibiotic derived from a microbial source. Cycloheximide is an antibiotic which is very active against many molds, yeasts, and phytopathogenic fungi. Cycloheximide (CHX) is an antibiotic produced by S. griseus. Its main biological activity is translation inhibition in eukaryotes resulting in cell growth arrest and cell death. CHX is widely used for selection of CHX-resistant strains of yeast and fungi, controlled inhibition of protein synthesis for detection of short-lived proteins and super-induction of protein expression, and apoptosis induction or facilitation of apoptosis induction by death receptors. Cycloheximide inhibits peptide synthesis in eukaryotic organisms but not in prokaryotes. Protein synthesis is blocked by the interaction of cycloheximide with the translocase enzyme. This interaction prohibits the translocation of messenger RNA on the cytosolic, 80S ribosomes without inhibiting organelle protein synthesis. Cycloheximide is also known to induce FAS/FAS Ligand apoptosis, and triggers apoptosis in HL-60 cells, T-cell hybridomas, Burkitt's lymphoma cells in addition to a variety of other cell types. Cycloheximide will also delay or inhibit apoptosis induced by other agents.
