The fragmentation of nuclear DNA is a hallmark of apoptotic cell death. The activities of caspase and nuclease are involved in the DNA fragmentation. Caspase-activated deoxyribo-nuclease (CAD), also termed DNA fragmentation factor (DFF40), is one such nuclease, and is capable of inducing DNA fragmentation and chromatin condensation after cleavage by caspase-3 of its inhibitor ICAD/DFF45. Caspase and CAD independent DNA fragmentation also exists. Recent studies demonstrated that another nuclease, endonuclease G (EndoG), is specifically activated by apoptotic stimuli and is able to induce nucleosomal fragmentation of DNA independently of caspase and DFF/CAD (1-3). EndoG is a mitochondrion-specific nuclease that translocates to the nucleus and cleaves chromatin DNA during apoptosis. The homologue of mammalian EndoG is the first mitochondrial protein identified to be involved in apoptosis in C. elegans (2). EndoG also cleaves DNA in vitro (4).

The integrin alpha 4 (also known as CD49d and ITGA4) belongs to the integrin alpha chain family of proteins. Integrins are heterodimeric integral membrane proteins composed of an alpha and beta chains (reviewed in 1). Alpha 4 (α4) chain associates with either beta 1 (β1) or beta 7 (β7) chain. It has been demonstrated that the putative ligand-binding sites of both integrin α4β1 and α4β7 is located on the α4 chain. These ligands included Madcam, VCAM, and fibronectin (2-4). Madcam is known as the principal ligand for integrin a4b7. Recently it was also demonstrated that HIV-1 envelope can mimic Madcam by binding to and signaling though integrin α4β7, the gut mucosal homing receptor for peripheral T cells (5). This recombinant protein fragment includes the ligand and monoclonal antibody binding domains; this binding has been tested and verified in ELISA assays. This recombinant protein is recognized PSI antibodies Integrin 4α (IN1) (PSI 4827) and Integrin 4α (IN2) (PSI 4837), but not Integrin 4α (CT) (PSI 4783).

Recent experiments have shown that IL-33 can also co-localize with heterochromatin and possesses transcriptional repressor activities, indicating that IL-33 may function as both a proinflammatory cytokine and an intracellular nuclear factor with transcriptional regulatory properties (5).

IL-16 was initially identified as a chemotactic cytokine (1), but is now known to possess a wide range of activities. Later studies have more fully characterized IL-16 as an immunomodulatory cytokine that contributes to the regulatory process of CD4+ T cell recruitment and activation at sites of inflammation in association with asthma and several autoimmune diseases (2). The precursor of IL-16 (pro-IL-16) is thought to be cleaved towards the C-terminal region by Caspase-3, releasing a 20 kDa active form that binds to and signals through CD4 (3,4). Besides acting as a chemotactic cytokine, IL-16 is thought to also be involved in the regulation of T cell proliferation (5) and multiple infectious, immune-mediated, and autoimmune inflammatory disorders including irritable bowel syndrome, systemic lupus erythematosus, and neurodegenerative disorders (6). At least two isoforms of IL-16 are known to exist; the longer isoform (also known as NIL-16) is detected only in neurons of the cerebellum and hippocampus (7). This recombinant protein fragment is derrived from this longer isoform.

MyD88 has been identified as an adapter molecule in the IL-1 signaling pathway (1). MyD88 associates with and recruits IRAK to the IL-1 receptor complex in response to IL-1 treatment and dominant negative form of MyD88 attenuates IL-1R-mediated NF-B activation. MyD88 is also employed as a regulator molecule by IL-18 receptor and human Toll receptor (2,3), which are members in the Toll/IL-1R family of receptors. Targeted disruption of the MyD88 gene results in lose of cellular responses to IL-1 and IL-18, and MyD88-deficient mice lack responses to bacterial product LPS that employs Toll-like receptors 2 and 4 (TLR2 and TLR4) as the signaling receptors. MyD88 is a general adapter protein for the Toll/IL-1R family of receptors and plays an important role in the inflammatory response induced by cytokines IL-1 and IL-18 and endotoxin. MyD88 gene is expressed in many tissues.

Influenza A virus is a major public health threat, killing more than 30,000 people per year in the USA (1). Novel influenza virus strains caused by genetic drift and viral recombination emerge periodically to which humans have little or no immunity, resulting in devastating pandemics. Influenza A can exist in a variety of animals, but it is in birds that all subtypes can be found (2). These subtypes are classified based on the combination of the virus coat glycoproteins hemagglutinin (HA) and neuraminidase (NA) subtypes. HA interacts with host cell surface proteins containing oligosaccharides with terminal sialyl residues. Its extracellular region has two domains (HA1 and HA2); HA1 is cleaved from the main hemagglutinin protein by the host immune system. During 1997, an H5N1 avian influenza virus was determined to be the cause of death in 6 of 18 infected patients in Hong Kong (3). This more recent virulent strain of H5N1 is now seen in Africa and Europe, as well as in Southeast Asia. There is some evidence of human to human spread of this virus, but it is thought that the efficiency of this type of transmission is low (4). Virus isolated from a human infected with the H5N1 strain in 1997 could bind to oligosaccharides from human as well as avian sources, indicating its species-jumping ability (5). This HA1 recombinant protein is recognized by several ProSci hemagglutinin antibodies.
