X box binding protein 1 (XBP-1) is a key protein in the mammalian unfolded protein response (UPR) that protects the cell against the stress of malfolded proteins in the endoplasmic reticulum (ER) (1). Upon sensing unfolded proteins, an ER transmembrane endonuclease and kinase termed IRE1p is activated and excises an intron from XBP-1 mRNA. The spliced XBP-1 mRNA results in a 371 amino acid protein (XBP-1s) which is then translocated to the nucleus where it binds to the regulatory elements of downstream genes. Together with other UPR transcription factors such as ATF6, XBP-1 stimulates the production of ER stress proteins including the ER resident protein chaperones glucose regulated protein (GRP) 78 and GRP94 (3,4).

In vertebrates, three main groups of actin isoforms, α, β and γ have been identified. The α actins are found in muscle tissues and are a major constituent of the contractile apparatus. The β and γ actins co-exist in most cell types as components of the cytoskeleton, and as mediators of internal cell motility. Actin, γ 1, encoded by this gene, is a cytoplasmic actin found in nonmuscle cells.
FUNCTION: Actins are highly conserved proteins that are involved in various types of cell motility and are ubiquitously expressed in all eukaryotic cells.

This process is negatively regulated by TOR (Target of rapamycin) through phosphorylation of autophagy protein APG1 (3). ATG5, another member of the autophagy protein family, forms a conjugate with ATG12; this conjugate has a ubiquitin-protein ligase (E3)-like activity for protein lipidation in autophagy (4). This conjugate also associates with innate immune response proteins such as RIG-I and VISA (also known as IPS-1), inhibiting type I interferon production and permitting viral replication in host cells (5).

Apoptosis plays a major role in normal organism development, tissue homeostasis, and removal of damaged cells. Disruption of this process has been implicated in a variety of diseases such as cancer (reviewed in 1). The Bcl-2 family of proteins is comprised of critical regulators of apoptosis that can be divided into two classes: those that inhibit apoptosis and those that promote cell death (reviewed in 2 and 3). MTCH2 is a member of the mitochondrial carrier protein family that catalyze the exchange of substrates across the inner mitochondrial membrane and is targeted by Bid, a pro-apoptotic Bcl-2 family member, in response to apoptotic signals (4-6), suggesting that MTCH2 may play a key role in the mitochondrial apoptosis pathway. The doublet observed in the SDS-PAGE is likely due to incomplete removal of the signal sequence.

Regulator of calcineurin 2 (RCAN2), also known as ZAKI4 and DSCR1L1, is expressed as two isoforms differing at their N-terminus. Apoptosis is related to many diseases and development. The p53 tumor-suppressor protein induces apoptosis through transcriptional activation of several genes. A novel p53 inducible pro-apoptotic gene was identified recently and designated PUMA (for p53 upregulated modulator of apoptosis) and bbc3 (for Bcl-2 binding component 3) in human and mouse (1-3). PUMA is one of the pro-apoptotic Bcl-2 family members including Bax and Noxa, which are also transcriptional targets of p53. The PUMA gene encodes two BH3 domain-containing proteins termed PUMA-α and PUMA-β (1). PUMA proteins bind Bcl-2, localize to the mitochondria, and induce cytochrome c release and apoptosis in response to p53. PUMA may be a direct mediator of p53-induced apoptosis. This PUMA recombinant protein is recognized by several ProSci PUMA monoclonal antibodies, but not the two polyclonal antibodies (PSI # 3041 and 3043).

The fragmentation of nuclear DNA is a hallmark of apoptotic cell death. The activities of caspase and nuclease are involved in the DNA fragmentation. Caspase-activated deoxyribo-nuclease (CAD), also termed DNA fragmentation factor (DFF40), is one such nuclease, and is capable of inducing DNA fragmentation and chromatin condensation after cleavage by caspase-3 of its inhibitor ICAD/DFF45. Caspase and CAD independent DNA fragmentation also exists. Recent studies demonstrated that another nuclease, endonuclease G (EndoG), is specifically activated by apoptotic stimuli and is able to induce nucleosomal fragmentation of DNA independently of caspase and DFF/CAD (1-3). EndoG is a mitochondrion-specific nuclease that translocates to the nucleus and cleaves chromatin DNA during apoptosis. The homologue of mammalian EndoG is the first mitochondrial protein identified to be involved in apoptosis in C. elegans (2). EndoG also cleaves DNA in vitro (4).
