Human Recombinant IL33 (from E. coli)

Interleukin-33 (IL-33) is a recently identified member of the IL-1 family of cytokines whose other members include IL-1, IL-1Ra and IL-18 (1,2). Its receptor has been shown to be ST2, an IL-1 receptor family member that also acts as a negative regulator of TLR-IL-1R signaling (1,3) and IL-1R accessory protein (IL-1RAcP) (4). Receptor binding of IL-33 activates NF-B and MAP kinases and induces the expression of TH2-associated cytokines such as IL-4, IL-5 and IL-6. Prolonged IL-33 treatment of mice led to the development of eosinophilia, splenomegaly, and severe pathological changes in mucosal organs such as lungs, esophagus and small intestine (2). Recent experiments have shown that IL-33 can also co-localize with heterochromatin and possesses transcriptional repressor activities, indicating that IL-33 may function as both a proinflammatory cytokine and an intracellular nuclear factor with transcriptional regulatory properties (5). This recombinant protein represents the cleaved and presumably activated form of IL-33, but has not been tested in any biological assays.

The pro-inflammatory cytokine IL-1 induced cellular response requires IL-1 receptor complex including IL-1RI and IL-1RAcP. MyD88 has been identified as an adapter molecule in the IL-1 signaling pathway (1). MyD88 associates with and recruits IRAK to the IL-1 receptor complex in response to IL-1 treatment and dominant negative form of MyD88 attenuates IL-1R-mediated NF-B activation. MyD88 is also employed as a regulator molecule by IL-18 receptor and human Toll receptor (2,3), which are members in the Toll/IL-1R family of receptors. Targeted disruption of the MyD88 gene results in lose of cellular responses to IL-1 and IL-18, and MyD88-deficient mice lack responses to bacterial product LPS that employs Toll-like receptors 2 and 4 (TLR2 and TLR4) as the signaling receptors. MyD88 is a general adapter protein for the Toll/IL-1R family of receptors and plays an important role in the inflammatory response induced by cytokines IL-1 and IL-18 and endotoxin. MyD88 gene is expressed in many tissues.

Influenza A virus is a major public health threat, killing more than 30,000 people per year in the USA (1). Novel influenza virus strains caused by genetic drift and viral recombination emerge periodically to which humans have little or no immunity, resulting in devastating pandemics. Influenza A can exist in a variety of animals, but it is in birds that all subtypes can be found (2). These subtypes are classified based on the combination of the virus coat glycoproteins hemagglutinin (HA) and neuraminidase (NA) subtypes. HA interacts with host cell surface proteins containing oligosaccharides with terminal sialyl residues. Its extracellular region has two domains (HA1 and HA2); HA1 is cleaved from the main hemagglutinin protein by the host immune system. During 1997, an H5N1 avian influenza virus was determined to be the cause of death in 6 of 18 infected patients in Hong Kong (3). This more recent virulent strain of H5N1 is now seen in Africa and Europe, as well as in Southeast Asia. There is some evidence of human to human spread of this virus, but it is thought that the efficiency of this type of transmission is low (4). Virus isolated from a human infected with the H5N1 strain in 1997 could bind to oligosaccharides from human as well as avian sources, indicating its species-jumping ability (5). This HA1 recombinant protein is recognized by several ProSci hemagglutinin antibodies.

PNGase F PRIME is a mutant recombinant PNGase F cloned from Flavobacterium meningosepticum and expressed and purified from E. coli. The proprietary changes made to PNGase F have been shown to have unique characteristics when compared to other commercially-available sources of PNGase F. Data generated by independent labs shows that PRIME works on native glycoproteins and serum glycoproteins in minutes at room temperature. Glycan analysis of the digestion products shows that PNGase F PRIME digestion led to more complete glycan release and also allowed for the cleavage of glycans not released by the commercially-available enzymes when used at the same concentrations with the same digestion conditions. This advancement benefits applications that seek to understand glycobiology in a natural milieu. When the immunoprecipitate is fully reduced immediately prior to SDS-gel electrophoresis, reactivity of Mouse IgG TrueBlot® with the 55 kDa heavy chains and the 23 kDa light chains of the immunoprecipitating antibody is minimized thereby eliminating interference by the heavy and light chains of the immunoprecipitating antibody in IP/Western blotting applications.
