Tumor Necrosis Factor Alpha (TNF alpha) is a protein secreted by lipopolysaccharide-stimulated macrophages, and causes tumor necrosis when injected into tumor bearing mice. TNF alpha is believed to mediate pathogenic shock and tissue injury associated with endotoxemia. TNF alpha exists as a multimer of two, three, or five non-covalently linked units, but shows a single 17kDa band following SDS PAGE under non-reducing conditions. TNF alpha is closely related to the 25kDa protein Tumor Necrosis Factor beta (lymphotoxin), sharing the same receptors and cellular actions. TNF alpha causes cytolysis of certain transformed cells, being synergistic with interferon gamma in its cytotoxicity. Although it has little effect on many cultured normal human cells, TNF alpha appears to be directly toxic to vascular endothelial cells. Other actions of TNF alpha include stimulating growth of human fibroblasts and other cell lines, activating polymorphonuclear neutrophils and osteoclasts, and induction of interleukin 1, prostaglandin E2 and collagenase production. TNF alpha is currently being evaluated in treatment of certain cancers and AIDS Related Complex.

Immunoglobulin gamma (IgG) is the most common class of antibody in blood and extracellular fluid. Approximately 75% of serum antibodies in humans are IgG. There are four immunoglobulin gamma subclasses: one, two, three and four. IgG1 is the most common, with 68% of all gamma class antibodies being G1, and G4 is the least common at 4%. Gamma class antibodies are found primarily in the secondary immune response, class switching from IgM and IgD. They are the only class of antibody that can cross the placenta, and along with IgA secreted in breast milk, provide the neonate with humoral immunity before immune system development occurs.

This antibody recognizes a protein of 75kDa identified as the gamma heavy chain of human immunoglobulins. It does not cross-react with alpha, mu, epsilon, or delta heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. The IgG antibody is useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. The most common feature of these malignancies is the restricted expression of a single heavy chain class. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is clonal and therefore malignant.

Lutropin (also designated luteinizing hormone) plays a role in spermatogenesis and ovulation by stimulating the testes and ovaries to produce steroids. Gonadotropin (also designated choriogonadotropin) production in the placenta maintains estrogen and progesterone levels during the first trimester of pregnancy. Ovaries and testes abundantly express luteinizing hormone/choriogonadotropin receptor. GnRH receptor contains seven hydrophobic transmembrane domains connected by hydrophilic extracellular and intracellular loops characteristic of G-protein coupled receptors. GnRH stimulates the gonadotrophs of the anterior pituitary to secrete luteinizing hormone (LH) as well as follicle-stimulating hormone (FSH). GnRH influences the protective effect of pregnancy and Gonadotropin against breast cancer. The expression of GnRH on breast carcinoma correlates in part to the degree of tumor differentiation. GnRH-positive breast tumors occur more frequently in tumors with greater cell differentiation in premenopausal women. GnRH is present in luteal and granulosa cells as well as in ovarian cell membrane preparations.

Deoxyribonuclease from beef pancreas, DNase I, was first crystallized by Kunitz. It is an endonuclease which splits phosphodiester linkages, preferentially adjacent to a pyrimidine nucleotide yielding 5'-phosphate terminated polynucleotides with a free hydroxyl group on position 3'. The average chain of limit digest is a tetranucleotide. DNase I acts upon single chain DNA, and upon double-stranded DNA and chromatin. In the latter case, although histones restrict susceptibility to nuclease action, over a period of time nearly all chromatin DNA is acted upon. According to Mirsky and Silverman, this could result from the looseness of histone attachment to DNA. They found that lysine-rich histones more effectively block DNase access to DNA than arginine-rich histones. Billing and Bonner suggest that DNase attacks the histone-free strand of chromatin DNA. Schmidt, et. al.indicate that hydrolysis of the histone-free region of DNA strands accounts for the initial rapid action of the enzyme on chromatin. Bollum reports degradation of synthetic homopolymer complexes by DNase I. The intracellular functions of the enzyme are probably controlled by a DNase inhibitor, which according to Lazarides and Lindberg is actin.

Cytokeratins, also called keratins or simply CKs, are a family of fibrous structural proteins and a major structural component in the outer layer of human skin, as well as hair and nails.  There are 20 human epithelial keratins which can be divided into two subfamiles: acidic and basic (more recent human genone sequencing has identified an additional 20 members). The formation of cytokeratin intermediate filaments requires the pairing of at least one acidic and one basic subfamily member. Members of the same keratin subfamily share extended sequence homology while members of different subfamilies show only limited sequence homology.

This antibody is specific for the 56.5kDa (CK10), 50kDa (CK14), 50kDa (CK15), 48kDa (CK16), and 40kDa (CK19) keratins of the acidic (Type I or LMW) subfamily. Many studies have shown the usefulness of CKs as markers in cancer research and tumor diagnosis, as epithelial tumors generally maintain the same CK expression patterns as their counterpart normal tissue. Clone AE1 is commonly used with clone AE3 as a pan cytokeratin antibody cocktail (Cat No V2330).

Tricine was first prepared by Good for use as a buffer for chloroplast reactions. It is structurally similar to Tris, but is much less inhibitory at high concentrations. For ATP assays using firefly luciferase, tricine buffer at 25 mM was found to be the best of ten common buffers tested.
Tricine is used as buffer component for separation of low molecular weight peptides. Tricine can be used in cryopreservation medium for the preservation of tissues and organs. Cryopreservation depends on the physical and chemical characteristics of the preservation medium used. The pH values and pK values for tricine/DMSO mixtures has been reported down to -20 °C. Tricine has been found to be an efficient scavenger of hydroxyl radicals in a study of radiation-induced membrane damage. Tricine is typically the buffer of choice in SDS-PAGE systems when separating proteins in the range of 1 to 100 kDa.
A buffer may be prepared by titrating with sodium hydroxide to the desired pH, using about a half-equivalent of NaOH.

Agarose is a purified linear galactan hydrocolloid isolated from agar or agar-bearing marine algae.
Agarose is useful in separation of nucleic acids electrophoretically because agarose gels have larger pore sizes than cross linked acrylamide gels at low concentrations and in chromatographic separations by either beading the agarose or cross-linking it. Agarose gels are also used in immunoelectrophoresis (IEP) and double-diffusion Ouchterlony plates to demonstrate antibody-antigen reactions. Agarose was used to prepare 0.5% agarose gel for Southern blot analysis of the DNA extracted from primate muscles.

Gel strength - the force that must be applied to a gel to cause it to fracture. Gel point - the temperature at which an aqueous agarose solution forms a gel as it cools. Electroendosmosis (EEO) - a movement of liquid through the gel. It occurs in living cells primarily in the oxidized state. Serves as a coenzyme of the dehydrogenases, especially in the dehydrogenation of primary and secondary alcohols. NAD usually acts as a hydrogen acceptor, forming NADH which then serves as a hydrogen donor in the respiratory chain.
Many metabolites and enzymes of biological interest are present in tissues at low concentrations. With the use of β-NAD as a catalyst intermediate and several enzymes in a multistep system, known as enzyme cycling, much greater sensitivity for detection of these components is achieved. The reduced form, β-NADH, is fluorescent whereas β-NAD is not. This difference in fluorescence provides a sensitive fluorescent measurement of the oxidized or reduced pyridine nucleotides at concentrations down to 10-7 M.

Citric acid is a key metabolic intermediate. Citrate is the starting point of the tricarboxylic acid cycle. Its concentration also coordinates several other metabolic pathways. Citric acid can form complexes with various cations, particularly with iron and calcium. In animals, citric acid improves the utilization of nutritional calcium. Citric acid is produced commercially by fermentation of carbohydrates derived from corn starch and from beet molasses.
Citric Acid, Trisodium Salt, Dihydrate is used as a substrate for citrate lyase, a buffer component; an anticoagulant. For anticoagulation use it is typically used at a concentration of approximately 0.129 M (i.e. for 4.5 mL blood use 16.0 mg sodium citrate and 2.1 mg citric acid).
To make a sodium citrate buffer use equimolar concentrations (typically approximately 0.05 M concentration) of citric acid, free acid and sodium citrate. Add equal volumes of each solution and titrate to the desired pH.

This mAb recognizes a protein of 134kDa, which binds fibroblast growth factor and E-selectin (cell-adhesion lectin on endothelial cells mediating the binding of neutrophils). Fucosylation is essential for binding to E-selectin. It contains sialic acid residues and 16 Cys-rich GLG1 repeats. This mAb can be used to stain the Golgi complex in cell or tissue preparations and can be used as a Golgi marker in subcellular fractions. It produces a diffuse staining pattern of the Golgi zone in normal and malignant cells. This mAb is an excellent marker for human cells in xenographic model research. It reacts specifically with human cells. The Golgi apparatus is an organelle present in all eukaryotic cells that forms a part of the endomembrane system. The primary function of the Golgi apparatus is to process and package macromolecules synthesized by the cell for exocytosis or use within the cell. The Golgi is made up of a stack of flattened, membrane-bound sacs known as cisternae, with three functional regions: the cis face, medial region and trans face.
