The NxGen® T4 DNA Ligases from LGC Biosearch (Lucigen) come in a range of highly pure grades specifically for next-generation sequencing (NGS). This enzyme catalyses the formation of a phosphodiester bond between the terminal 5′ phosphate and 3′ hydroxyl groups of duplex DNA or RNA and efficiently joins blunt and cohesive ends, and repairs single stranded nicks in duplex DNA, RNA, or DNA/RNA hybrids.

10X T4 DNA Ligase Buffer is provided with T4 DNA Low concentration ligase.
2X Rapid Ligation Buffer and 10X T4 DNA Ligase Buffer is provided with T4 DNA High concentration ligase.

10X T4 DNA Ligase Buffer is composed of 500mM Tris-HCI, 100 mM MgCl2, 50 mM dithiothreitol, 10 mM ATP, pH 7.6 @ 25°C.
2X Rapid Ligation Buffer is composed of 132mM Tris-HCI, 20 mM MgCl2, 2 mM dithiothreitol, 2 mM ATP, 15% PEG, pH 7.6 @ 25°C.

Performance Specifications: 1) SS Exonuclease: 6,000 U <0.1% released. 2) DS Exonuclease: 6,000 U <0.1% released. 3) Endonuclease: 6,000 U <0.1% converted. 4) E. coli 16S rDNA Contamination: 3,000 U < 10 copies. 5) Storage: -20°C.

Ordering information: Enzyme is provided in 10 mM Tris-HCl, 50 mM NaCl, 1 mM dithiothreitol, 0.1 mM EDTA, 50% glycerol, pH 7.5 at 25°C. Unit definition: 1 unit is defined as the amount of T4 DNA Ligase required to join 50% of 100 ng of DNA fragments with cohesive termini in 50 µl 1 X T4 DNA Ligase Buffer following a 30 minute incubation at 23°C.
