The deduced NSHC sequence encodes isoforms of 474 and 594 amino acids, corresponding to proteins of 52 and 64 kD, respectively. Sequence analysis determined that NSHC shares an overall homology of 54% with SHC and approximately 70% homology in the PTB and SH2 domains. Northern blot analysis detected a 10-kb NSHC transcript exclusively in brain, whereas SHC was expressed as 3.8-, 3.4-, and 2.8-kb transcripts in all tissues except brain. Within the central nervous system, NSHC was highly expressed in cerebral cortex, hippocampus, amygdala, and caudate nucleus, with low-level expression in subthalamic nucleus, thalamus, putamen, corpus callosum, hypothalamus, medulla, substantia nigra, and cerebellum. Expression of NSHC was undetectable in the spinal cord.
