About this item

Recombinant human PDGFR alpha (D842V) (550-end) point mutant was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag. PDGFR alpha is a member of the PDGFR family of membrane receptors with intrinsic tyrosine kinase activity.

  • Includes kinase, substrate and reaction buffer
  • Use with ADP-Glo™ Assay for bioluminescent detection of kinase activity

Recombinant human PDGFR alpha (D842V) (550-end) point mutant was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag. PDGFR alpha (platelet-derived growth factor receptor alpha) is a member of the PDGFR family of membrane receptors with intrinsic tyrosine kinase activity. Aberrant expression of PDGFR alpha has been linked to developmental abnormalities in vertebrate models and implicated in multiple disease states in humans. There is widespread expression of PDGFR alpha in renal cell types involved in fibrotic and sclerosing processes (1). PDGF and its receptor PDGFR alpha are inducers of fibrosis in the repair phase of inflammatory bowel disease, and they may also be involved in the active inflammatory phase (2). ADP-Glo Kinase Assay is a luminescent kinase assay that measures ADP formed from a kinase reaction; ADP is converted into ATP, which is a substrate in a reaction catalyzed by Ultra-Glo Luciferase that produces light. The luminescent signal positively correlates with ADP amount and kinase activity. The assay is well suited for measuring the effects chemical compounds have on the activity of a broad range of purified kinases, making it ideal for both primary screening as well as kinase selectivity profiling. The ADP-Glo Kinase Assay can be used to monitor the activity of virtually any ADP-generating enzyme (e.g., kinase or ATPase) using up to 1mM ATP. Kinase Enzyme System contains: Kinase: PDGFR alpha (D842V), 10ug (Human, recombinant; amino acids 550-end). MW: ~95kDa. Substrate: Native Swine Myelin Basic Protein (MBP). Other: Reaction Buffer, DTT, MnCl2. PDGFR alpha (D842V) NCBI Database Entry: www.ncbi.nlm.nih.gov/gene/5156/. Visit www.promega.com/kinase/ to see all Kinase Enzyme Systems.
