Recombinant full-length human CDK5 and human p35 were co-expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag. CDK5 is a member of the Cyclin-Dependent Kinase family that is most abundant in the mammalian brain. The active form of CDK5, which has also been called neuronal cdc2-like kinase, is a heterodimer of CDK5 and a 25kDa protein, which is derived proteolytically from a 35kDa brain and neuron-specific protein and is essential for the kinase activity of CDK5. CDK5 has emerged as a crucial regulator of neuronal migration in the developing central nervous system. CDK5 phosphorylates a diverse list of substrates, implicating it in the regulation of a range of cellular processes---from adhesion and motility to synaptic plasticity and drug addiction. ADP-Glo Kinase Assay is a luminescent kinase assay that measures ADP formed from a kinase reaction; ADP is converted into ATP, which is a substrate in a reaction catalyzed by Ultra-Glo Luciferase that produces light. The luminescent signal positively correlates with ADP amount and kinase activity. The assay is well suited for measuring the effects chemical compounds have on the activity of a broad range of purified kinases, making it ideal for both primary screening as well as kinase selectivity profiling. The ADP-Glo Kinase Assay can be used to monitor the activity of virtually any ADP-generating enzyme (e.g., kinase or ATPase) using up to 1mM ATP.
