Zymolyase-100T is an enzyme preparation from a submerged culture of Arthrobacter luteus which effectively lyses cell walls of viable yeast cells.

- Primary enzyme responsible for lysis of viable yeast cells in ß-1,3-glucan laminaripentaohydrolase
- Hydrolyzes glucose polymers at the ß-1,3-glucan linkages releasing laminaripentaose as the principal product
Zymolyase 100T is used to lyse living yeast cell walls to produce protoplast or spheroplast of various strains of yeast cells.
The lytic spectrum of this enzyme includes organisms from the following genera: Ashbya, Candida, Debaryomyces, Eremothecium, Endomyces, Hansenula, Hanseniaspora, Kloekera, Kluyveromyces, Lipomyces, Metschikowia, Pichia, Pullularia, Torulopsis, Saccharomyces, Saccharomcopsis, Saccharomycodes, Schwannimomyces and others.
Activators: Dithiothreitol, 2-mercaptoethanol and other sulfhydryl compounds (such as cysteine); Essential Enzyme: ß-1,3-glucan laminaripentaohydrolase; Other Enzymes: ß-1,3-glucanase, ca. 1.5×107 units/g, Protease, ca. 1.0×104 units/g, Mannase, ca. 1.0×104 units/g.
Zymolase-100T has a low solubility. Use as a suspension. If it is necessary to make a sterile enzyme solution of more than 0.05%, make a 2% enzyme stock solution by dissolving Zymolase -100T in a buffer solution (pH 7.5) which contains 5% glucose. To remove insoluble substances, filter through a sterile filter. Do not use Nitrocellulose filters. Dilute sterile enzyme suspension to desired concentration.
It may also be used for glucan hydrolysis.
