The Cathepsin Activity Assay Kits are based on fluorometric or colorimetric (Cathepsin G assay) detection and utilize the preferred substrate sequence for each cathepsin, labeled with AFC (amino-4-trifluoromethylcoumarin), MCA (7-amino-4-methyl coumarin) or pNA (p-nitroaniline). Cell lysates and other samples that contain cathepsins cleave the synthetic substrate to release free AFC, MCA or pNA that can be easily quantified using a fluorometer/photometer or a fluorescence plate reader/ELISA reader. The cathepsin assays are easy to perform and can be adapted to 96-well plate assays. Assay conditons have been optimized to obtain the maximal activity.
