The standard working range is 100-1500 µg/mL with up to 1% detergent (or higher in some cases). Protein concentrations are estimated by reference to absorbances obtained for a series of standard protein dilutions, typically bovine serum albumin (BSA), which are assayed alongside the unknown samples. Because the color response with coomassie dye is non-linear with increasing protein concentration, a standard curve must be completed with each assay. Standards can be used directly without preparing them in the same detergent found in the test samples.
