Human peripheral blood macrophages (mixed) is a 50%-50% mixture of M1 and M2 macrophages after harvest
PBMCs are collected in HemaCare's FDA-registered collection centers following cGMP and cGTP collection guidelines from healthy human volunteer donors under IRB approved informed consent by leukapheresis using the COBE® Spectra Apheresis System MNC collection protocol. This mononuclear cell-rich leukapheresis product (LeukoPak) is extracted using continuous flow centrifugal technology directly into the collection bag. Mononuclear cells are purified using density gradient centrifugation techniques.
Serological and Infectious disease testing are performed with each donation. Customers should handle product as if potentially infectious following biosafety level 2 procedures.
M1 and M2 macrophages are derived from purified monocytes. CD14 monocytes were isolated from peripheral blood mononuclear cells (PBMCs) using positive immunomagnetic cell separation procedures. M1 macrophages are cultured in the presence of GM-CSF for 10 days and M2 macrophages are cultured in the presence of M-CSF for 10 days. After 10 days, macrophages are combined at a 50%-50% ratio.
