Human Recombinant IFN-alpha 2B
Supplier: STEMCELL Technologies
Interferon-alpha (IFN-α) is a type I interferon, produced by virus infected cells, and is released as a soluble factor to initiate antiviral responses (Isaacs and Lindenmann). IFN-α2 is the most potent IFN-α used in fundamental research and in most clinical applications. The best known IFN-α2 subvariants, 2A and 2B, differ by only one or two amino acids at positions 23 and/or 34 of the mature protein (von Gabain et al.). Type I IFNs exert potent antitumor activity by increasing the cytotoxic activity of NK and T cells, as well as inhibiting the proliferation of cancer cells (Paul et al.). Additionally, it has been shown that proinflammatory IFN-α modulates the function of B cells in patients with systemic lupus erythematosus (Chang et al.) and pegylated form of IFN-α 2A and 2B has implications in the treatment of Hepatitis C (Foster et al.).

Human Recombinant EGFR
Supplier: STEMCELL Technologies
Epidermal growth factor receptor (EGFR) is a type I transmembrane protein and receptor tyrosine kinase. EGFR has been shown to bind to some members of the EGF family ligands including EGF, amphiregulin, TGF-α, betacellulin, epiregulin, heparin-binding EGF, and neuregulin-2α. EGFR ligand binding induces homodimerization, as well as heterodimerization of EGFR with ErbB2 or with ligand-activated ErbB3 or ErbB4 (Schlessinger). Dimerization results in kinase activation, phosphorylation, and cell signaling, mediated primarily through MEK/ERF and AKT pathways (Navlonic et al.). EGFR signaling has been shown to regulate cell proliferation, differentiation, motility, and apoptosis. Elevated levels of EGFR have been correlated with carcinogenesis (Maihle et al.). Protein contains a His-residue tag at the carboxyl end of the polypeptide chain.

Mouse Recombinant MIP-1 alpha (CCL3)
Supplier: STEMCELL Technologies
Macrophage inflammatory protein-1 alpha (MIP-1 alpha), also known as CCL3, is a member of the CC family of chemokines and is most closely related to CCL4 or MIP-1 beta. Mouse MIP-1 alpha signal through CCR1, CCR3, CCR5, and D6 receptors (Menten et al.). MIP-1 alpha exhibits a variety of proinflammatory activities in vitro, including leukocyte chemotaxis, cytokine production, and mast cell activation, and it inhibits the proliferation of hematopoietic stem cells in vitro and in vivo (Cook). MIP-1 alpha plays a critical role in macrophage recruitment into wounds and in tissue repair (DiPietro et al.). It has been demonstrated that blockade of the CCL3/MIP-1 alpha-CCR1 pathway blocks the recruitment of CCR1-expressing CD4+ T cells to the liver, showing a therapeutic potential for treating T cell-mediated liver diseases (Ajuebor et al.).

Mouse Recombinant IL-1 alpha
Supplier: STEMCELL Technologies
Interleukin 1 alpha (IL-1α) is a member of the IL-1 family and a dual-function cytokine. Both the unprocessed precursor and a processed IL-1α protein signal through IL-1 receptor type 1 (IL-1R1). Various cells, including keratinocytes, thymic epithelium, hepatocytes, endothelial cells, fibroblasts, and the epithelial cells of mucous membranes have high levels of intracellular IL-1α precursor, which is also expressed on the surface of monocytes and B lymphocytes (Netea et al.). IL-1α recruits infiltrating cells to a site of injury during necrosis and plays an important role during processes of sterile inflammation (Rider et al.; Cohen et al.). During hypoxia, IL-1α contributes to angiogenesis (Carmi et al.). IL-1α is produced by microglia-like cells after ischemic brain injury, which contributes to the inflammation (Luheshi et al.).

Human Recombinant IL-3 (CHO-expressed)
Supplier: STEMCELL Technologies
Interleukin 3 (IL-3) is a species-specific pleiotropic cytokine that promotes the survival and proliferation of pluripotent hematopoietic stem cells and lineage-committed progenitor cells and their differentiation into mature cells of most lineages, including basophils, neutrophils, eosinophils, macrophages, dendritic cells, erythrocytes, and megakaryocytes (Yang et al.; Dorssers et al.; Broughton et al.). IL-3 is produced by activated T cells and has a physiological role in inflammation and allergies by promoting the secretion of inflammatory mediators such as histamine, IL-4, and IL-6 by basophils and eosinophils (Broughton et al.). The IL-3 receptor consists of a unique alpha subunit (CD123) and a beta common subunit (βc or CD131) that is shared with the receptors for IL-5 and GM-CSF, and is the principal signal transduction subunit for these cytokines. IL-3 binding to the heterodimeric receptor activates JAK/STAT, MAPK, and PI3K signaling pathways (Woodcock et al.).

Human Recombinant Persephin, ACF
Supplier: STEMCELL Technologies
Persephin is a neurotrophic factor that belongs to the glial cell line-derived neurotrophic factor (GDNF) family. Persephin shares a large degree of structural similarity to GDNF, artemin, and neurturin, and has overall neuroprotective activity. Persephin signals through GRFα4 (glycosylphosphatidylinositol (GPI)-linked GDNF receptor family member) which signals through the receptor tyrosine kinase RET. Unlike GDNF and neurturin, persephin only promotes the growth and survival of central dopaminergic and motor neurons, but not peripheral neurons (Milbrandt et al.). In vitro persephin only promotes survival of neurons that co-express GPI-linked GRFα4 and RET (Enokido et al.; Lindahl et al.). Mice lacking persephin showed increased cell death after cerebral ischemia, however administration of persephin before ischemia dramatically reduced neuronal cell death (Tomac et al.). This product is animal component-free.

VZV (IE63) Peptide Pool
Supplier: STEMCELL Technologies
Stimulating T cells with VZV (IE63) peptide pool releases downstream cytokines and upregulates activation markers, enabling antigen-specific T cells to be detected or isolated for analysis. VZV (IE63) peptide pool is a lyophilised mixture of 67 peptides from the transcriptional regulator ICP22 homolog (IE63) of Varicella-zoster virus (VZV; strain Oka vaccine), and consists of 15-mer peptides with 11-amino-acid overlaps that cover amino acids 1 to 278 on IE63. Expressed during VZV latency and reactivation (Lungu et al.), IE63 demonstrates repressive activity in both the cytoplasm and nucleus of infected cells (Bontems et al.). Host interferon alpha (IFN-α) activity is inhibited by VZV IE63, which likely contributes to VZV pathogenesis (Ambagala and Cohen). Viral peptide pools are useful for a broad range of applications, including vaccine development, immunological research, and diagnostic assay development.

Human Recombinant gAdiponectin (gAcrp30), ACF
Supplier: STEMCELL Technologies
gAdiponectin, also known as an adipocyte component-related protein of 30 kDa, is a member of defense collagens, and its structure is similar to C1q-a member of the complement-related family of proteins (Chandran et al.). gAdiponectin is expressed by differentiated adipocytes and it is composed of three defined domains: N-terminus-containing hypervariable region, collagenous stalk containing 22 GXY repeats, followed by a C-terminus globular domain (Wang et al.). gAdiponectin has also been identified as a hematopoietic stem cell growth factor (DiMascio et al.), and is known to regulate bone marrow mesenchymal stem cell niche (Yu et al.). The globular domain of adiponectin (gAcrp30) has been shown to significantly reduce plasma-free fatty acids and glucose levels in mice (Wong et al.), and to regulate glucose and fatty acid uptake and oxidation in cardiomyocytes (Palanivel et al.). This product is animal component-free.

Human Recombinant NGF-beta, ACF
Supplier: STEMCELL Technologies
Nerve growth factor (NGF)-beta is a prototypical member of the neurotrophin family and has a role in the survival and growth of neural cells, regulating cell growth, promoting differentiation into neurons, and neuron migration. The beta subtype of NGF is biologically active in comparison to the alpha-2 and gamma-2 subtypes. NGF-beta in its secreted form can bind to tyrosine kinase A (trkA) receptor with high affinity and to p75 (NTR) with low affinity (Levi and Alemà; Sofroniew et al.). NGF has been shown to possess pro-inflammatory and pro-fibrogenic properties (Micera et al.). It has also been shown that overexpression of NGF-beta promotes differentiation of bone marrow mesenchymal stem cells into neurons through regulation of AKT and MAPK pathways (Yuan et al.). This product is animal component-free.

Mouse Recombinant IL-3
Supplier: STEMCELL Technologies
Interleukin 3 (IL-3) is a species-specific pleiotropic cytokine that promotes the survival and proliferation of pluripotent hematopoietic stem cells and lineage-committed progenitor cells and their differentiation into mature cells of most lineages, including mast cells, basophils, neutrophils, eosinophils, macrophages, dendritic cells, erythrocytes, and megakaryocytes (Fung et al.; Metcalf et al.; Broughton et al.). IL-3 is produced by activated T cells, and has a physiological role in inflammation and allergies by promoting the secretion of inflammatory mediators such as histamine, IL-4, and IL-6 by mast cells, basophils and eosinophils. The mouse IL-3 receptor consists of a unique alpha-subunit (CD123) and two beta subunits, one specific for IL-3 (βIL-3), the other shared with the receptors for IL-5 and GM-CSF (beta common chain, βc or CD131). IL-3 binding to heterodimeric receptors containing the alpha subunit and one of either beta subunits activates JAK/STAT, MAPK, and PI3K signaling pathways (Scott and Begley).

Human Recombinant TNF-alpha, ACF
Supplier: STEMCELL Technologies
Tumor necrosis factor-α (TNF-α) is a pro-inflammatory cytokine that activates NF-kB, MAPK, and PI3K/AKT pathways. Activated T cells and macrophages are the primary producers of TNF-α in response to inflammation and infectious conditions. Many other cell types have been shown to produce TNF-α, among them B cells, NK cells, mast cells, neutrophils, dendritic cells, microglia, endothelial cells, smooth muscle cells, cardiomyocytes, and fibroblasts. TNF-α has cytotoxic effects on cancer cells in vitro by stimulating anti-tumor immuno- suppressive responses. TNF-α stimulates expression of E- and P-selectins, thus facilitating adhesion of neutrophils, monocytes, and memory T cells to activated platelets and endothelial cells (Zelová and Hosek). Other effects of TNF-α include vasodilatation and edema formation. This product is animal component-free.

Human Recombinant FGF-7 (KGF)
Supplier: STEMCELL Technologies
Fibroblast growth factor 7 (FGF-7) is a member of the FGF family, and acts exclusively through a subset of FGF receptor isoforms expressed predominantly by epithelial cells (Finch and Rubin). FGF-7 seems to act specifically on epithelial cells and stimulates proliferation, migration, and differentiation of these cells, and also participates in epithelial protection and repair both in vitro and in vivo (Finch and Rubin; Werner). In contrast, FGF-7 is produced solely by cells of mesenchymal origin, and functions as a paracrine mediator of mesenchymal-epithelial communication (Rubin et al.). FGF-7 has also been shown to supplement several wound-healing properties of bioengineered skin (Erdag et al.) and to induce autophagy in human keratinocytes (Belleudi et al.). Additionally, FGF-7 has a role in pluripotent stem cell differentiation to endodermal pancreatic-like insulin-producing cells and thymic epithelial cells (Inami et al.; Niu et al.).

HPV16 (E6) Peptide Pool
Supplier: STEMCELL Technologies
Stimulating T cells with HPV16 (E6) Peptide Pool releases downstream cytokines and upregulates activation markers, enabling antigen-specific T cells to be detected or isolated for analysis. HPV16 (E6) Peptide Pool is a lyophilised mixture of 37 peptides from the E6 protein of human papillomavirus type 16 (HPV16). The pool consists of 15-mer peptides with 11-amino-acid overlaps that cover amino acids 1 to 158 on protein E6. E6 binds to the tumor suppressor protein p53 and stimulates its degradation (Scheffner et al.; Werness et al. ), which is thought to cause an increase in spontaneous mutagenesis in host cells (Havre et al.). E6 together with E7 demonstrated the ability to immortalize human keratinocytes in vitro, which likely has significance for HPVs oncogenic potential (Hawley-Nelson et al.). Viral peptide pools are useful for a broad range of applications, including vaccine development, immunological research, and diagnostic assay development.

Mouse Recombinant FGF-21
Supplier: STEMCELL Technologies
Fibroblast growth factor 21 (FGF-21) is a member of the FGF family. Using β-Klotho as a cofactor, FGF-21 signals through FGF receptor 1c and 4 to activate PI3K and MAPK pathways (Mattila and Härkönen; Kharitonenkov et al.). FGF-21 expression is regulated by tissue-specific peroxisome proliferator-activated receptors (PPARs). Upon PPAR-α stimulation FGF-21 is produced in the liver, and activation of PPAR-γ leads to FGF-21 production in adipose tissue. FGF-21 promotes insulin-independent glucose uptake and lipid accumulation in primary human adipocytes and in mouse 3T3-L1 cells. In pancreatic islets and INS-1 cells it inhibits glucose-mediated glucagon release and stimulates insulin production. FGF-21 does not induce proliferation in immortalized cell lines, unlike other FGFs (Kharitonenkov and Shanafelt). FGF-21 regulates thermogenesis in white and brown adipose tissue, and metabolic processes in cells of pancreatic origin (Kharitonenkov et al.).
