Poly-lysine is a polycation which binds to DNA, red cell membrane and any negatively charged protein. When adsorbed to the culture surface, poly-lysine increases the number of positively charged sites available for cell binding. A compromise between the easier to use lower molecular weight products and the extremely viscous higher molecular weights would be the products in the range of 70,000-150,000.
It is typically used as a coating substrate for culture dishes, slides, etc. It enhances electrostatic interaction between negatively charged ions of the cell membrane and the culture surface. Both the D- and L- form of the poly-lysine can be used as a coating substrate since poly-lysine is a nonspecific attachment factor for cells; however, certain cells can digest poly-lysine. In this case, poly-D-lysine should be used as the attachment factor so that the cells are not disrupted by excessive uptake of L-lysine.
Other uses for poly-lysine have been reported as well:
• Conjugation to methotrexate for increased drug transport.
• Microencapsulation of islets.
• Use in simple reproducible procedure for chromosomal preparations from a variety of tissues.
• Immobilation of living cells in biocompatible semipermeable microcapsules.
• In the preparation of polycationic beads.
• Conjugation to albumin and horseradish peroxidase to enhance cellular uptake.
Poly-D-lysine hydrobromide is a synthetic amino acid that enhances cell adhesion to solid substrates. It has additionally been shown to eliminate prion proteins from infected cells.

Recently, a number of cytokines belonging to the interleukin (IL)-17 family have been identified. These are termed as IL-17B, IL-17C and IL-17E. IL-17 is a potent proinflammatory cytokine that plays roles in a number of diseases including rheumatoid arthritis , multiple sclerosis , and promotion of tumor growth. IL-17B, C, and E like IL-17 are able to induce proinflammatory responses. However, they do not bind to the IL-17 receptor suggesting that additional IL-17R related receptor might exist. Receptor for IL-17B and IL-17E has been independently isolated by Shi, et al and Lee, et al. and has been designated as EV127 (in mouse) and IL-17Rh1 (in human), respectively. IL-17E induces activation of NF-κB pathway and like IL-17 also induces production of IL-8. The IL17 proteins are a family of potent cytokines that act to induce proinflammatory responses. Studies have shown that IL17E binds strongly to IL17RB. Receptor binding of ligand has been shown to lead to the activation of nuclear factor kappa-B and production of IL8. Exposure of mice to IL17 resulted in a Th-2 like response characterized by increased serum IgE, IgG1 and IgA levels, blood eosinophilia, increased lymphocytes and neutrophils, and pathological changes in the tissues that included eosinophilic infiltrates, increased mucus production, B-lymphocyte hyperplasia and epithelial cell hyperplasia/hypertrophy.

Glutathione is the major low molecular weight thiol compound of the living plant or animal cell. It is a tripeptide with a gamma peptide linkage between the amine group of cysteine (which is attached by normal peptide linkage to a glycine) and the carboxyl group of the glutamate side-chain. It is an antioxidant, preventing damage to important cellular components caused by reactive oxygen species such as free radicals and peroxides. The sulfhydryl (thiol) group (SH) of cysteine serves as a proton donor and is responsible for the biological activity of glutathione.
Glutathione suppresses human immunodeficiency virus expression in chronically infected monocytic cells. It is a useful tripeptide involved in many aspects of metabolism, including transport of g-glutanyl amino acids and reductive cleavage of disulfide bonds.
Endogenous antioxidant that plays a major role in reducing reactive oxygen species formed during cellular metabolism and the respiratory burst. Glutathione-S-transferase catalyzes the formation of glutathione thioethers with xenobiotics, leukotrienes, and other molecules that have an electrophilic center. Glutathione also forms disulfide bonds with cysteine residues in proteins. Via these mechanisms, it can have the paradoxical effect of reducing the efficacy of anti-cancer agents.

Macrophage colony-stimulating factor (M-CSF) is a homodimeric cytokine that belongs to receptor tyrosine kinase subclass III of the receptor tyrosine kinase (RTK) family. M-CSF acts on a CSF-1 receptor tyrosine kinase, which initiates signaling cascades to support cell proliferation and differentiation (Hamilton). M-CSF is produced by endothelial cells, osteoblasts, and during pregnancy by the uterine epithelial cells (Ryan). M-CSF synergizes with other factors to support proliferation and differentiation of multipotent hematopoietic progenitor cells, and regulates proliferation and differentiation of the mononuclear phagocyte progenitor cells to monocytes and macrophages. It also supports survival, proliferation, and function of the differentiated macrophages and regulates differentiation of mononuclear phagocytes to osteoclasts (Pixley and Stanley). M-CSF plays an important role in the implantation of the embryo and early development (Makrigiannakis et al.).

Interleukin 12 (IL-12p70) is a heterodimeric cytokine composed of p35 and p40 subunits. IL-12 is produced by monocytes, macrophages, dendritic cells, neutrophils, and B cells in response to bacterial products and cytokines such as IFN-γ. The IL-12 receptor is expressed on T, NK, and dendritic cells. Upon binding, IL-12 initiates signaling via the JAK/STAT signaling pathway and stimulates NK, B, and T cells to produce IFN-γ (Watford et al.). It also regulates cytokine synthesis, proliferation of T and NK cells, and stimulates differentiation of CD4+ and CD8+ T cells (Germann and Rüde). Mice that are deficient in IL-12 are susceptible to many intracellular pathogens and have impaired IFN-γ secretion, Th1 differentiation, and NK cytolytic activity; however, Th2 development and IL-4 production are enhanced (Watford et al.).

Fibroblast growth factor 4 (FGF-4) is a member of the FGF superfamily (Beenken and Mohammadi). FGF-4 is expressed in pluripotent stem cells and is implicated in various stages of development and morphogenesis in a variety of organisms (Kosaka et al. 2009). FGF-4 has been shown to have an ability to promote neural stem cell proliferation and neuronal differentiation in the postnatal brain (Kosaka et al. 2006). FGF-4 has also been shown to increase the proliferation rate of mesenchymal stem cells (Farré et al.). FGF-4 supports the maintenance and self-renewal properties of human embryonic stem cells and also promotes the proliferation of these cells (Mayshar et al.). In the mouse, the Fgf4 gene also supports proliferation of the inner cell mass (ICM) and postimplantation embryos (Feldman et al.).

Lupeol, a dietary triterpene found in certain fruits, vegetables, and medicinal plants, has potent anti-inflammatory, anticarcinogenic, antimutagenic, and antimalarial activity. It suppresses the growth of hepatocellular carcinoma cell lines SMMC7721 and HepG2 with IC50 values of 45 and 48.5 μM and melanoma cell lines Mel 928 and Mel 1241 with IC50 values of 75 and 72 μM.¹'² At 0.76 g/kg lupeol causes a significant decrease in the blood pressure of stroke-prone hypertensive rats and reduces expression of hepatic genes involved in triglyceride and cholesterol synthesis.³ It also has significant anti-inflammatory effects at 50 mg/kg in a carrageenan-induced edema model, inhibiting neutrophil migration.⁴
