Exonuclease III

Exonuclease III, the major apurinic/apyramadinic (AP) endonuclease in E. coli, is a 3' to 5' exonuclease digesting one strand of a dsDNA duplex at a time or digesting the RNA strand of an RNA-DNA heteroduplex. Exonuclease III breaks phosphodiester bonds on the 5' side of AP sites in both dsDNA and ssDNA, removes 3'mterminal groups on dsDNA, increases MutY turnover, and efficiently degrades 3' recessed but not 3' protruding DNA ends (creating 5' overhangs). These properties have been exploited to allow strand specific labelling, unidirectional deletions, and selective strand degradation.

RNase I

T4 Endonuclease VII is a DNA junction specific endonuclease (also referred to as a resolvase or a cleavase). T4 Endonuclease VII is the product of gene 49 of phage T4. This gene product has 157 amino acid residues and a mass of 18 kDa. T4 Endonuclease VII functions in vivo to resolve branched DNA structures in newly synthesised DNA. It does this by cleaving the DNA flanking the branch prior to rearrangement and reannealing of the DNA strands.

Shrimp recombinant Dnase

Shrimp DNase is an endonuclease that cleaves phosphodiester linkages in DNA to yield di- and oligonucleotides with 5’-phosphate and 3’-hydroxyl termini. This DNase has a remarkably high specific activity towards double-stranded DNA (dsDNA). The activity towards dsDNA is 5000-fold higher than towards single-stranded DNA, and thus can be used selectively to degrade dsDNA, leaving single-stranded DNA intact. The activity of this enzyme depends on Mg²⁺ concentration and is stimulated by Ca²⁺. However, Ca²⁺ also stimulates the RNase activity of shrimp DNase and should be avoided when RNA integrity is critical.

DNase I

DNase I is an endonuclease that catalyses the degradation of both single and double-stranded DNA to yield di-, tri-, and oligonucleotides with 5'-phosphate and 3'-hydroxyl termini. DNase I is stabilised by the addition of Ca²+ and requires a divalent metal cation for activity. It randomly produces nicks in double-stranded DNA in the presence of Mg²+, but in the presence of Mn²+, both strands of double-stranded DNA are cleaved leaving blunt-end fragments or those with one or two protruding nucleotides.
