About this item

OPA1-03142 detects human phosphorylated Tau from human samples. This antibody is expected to cross react with bovine, goat, baboon, monkey (100%), mouse, and rat (92%) based on sequence homology. OPA1-03142 has been successfully used in Western blot procedures. The OPA1-03142 immunogen is a synthetic phosphopeptide that was derived from a region of mouse tau that contains serine 262. Tau is a neuronal microtubule-associated protein found predominantly on axons. The function of Tau is to promote tubulin polymerization and stabilize microtubules. Tau, in its hyper-phosphorylated form, is the majoy component of paried helical filaments (PHF), the building block of neurofibrillary lesions in Alzheimer's diseases (AD) brain. Hyper-phosphorylated Tau is also found in neurofibrillary lesions in a range of other central nevous system disorders. Hyperphosphorylation impairs the microtubule binding function of Tau, resulting in the destabilization of microtubules in AD brains, ultimately leading to the degeneration of the affected neurons. Numerous serine/threonine kinases, including GSK-3beta, protein kinase A (PKA), cyclindependent kinase 5 (cdk5) and casein kinase II, phosphorylate Tau. Serine 262 can be phosphorylated by GSK-3beta, PKA, CamKII and MARK, and has been found to be a major site in AD brain. This antibody, combined with Tau [pS^356] antibody, enables one to distinguish between phosphorylation at the two distinct residues, rather then a combined epitope as detected by 12E8 antibody.
