NOV, also called CCN3, is a secreted protein of CCN family members. CCN family members are highly conserved cysteine rich proteins sharing a common modular structure having 4 conserved domains, insulin-like growth factor-binding protein (IGFBP) domain, von Willebrand type C (VWC) domain, thrombospondin-1 (TSP-1) domain, and C-terminal (CT) domain (absent in CCN5). By specific interactions with these domains, CCN proteins modulate multiple signalling pathways including BMPs, Wnt, TGFs, Notch and integrins to regulate cell proliferation, differentiation, adhesion, migration, angiogenesis, and survival. CCN3 is firstly characterised as a promoter of progenitor activity of human hematopoietic stem cells, as knockdown of CCN3 can abrogate the function of primitive progenitors. Recent studies showed that CCN3 is also actively involved in the process of wound healing. CCN3 is highly expressed in granulation tissues of cutaneous wounds and capable of inducing synthetic responses of fibroblasts.

IKAP Antibody: IKAP was initially identified as a scaffold protein of the I kappa B kinase complex that could bind to IKK alpha , IKK beta , NF-kappa B, and the NF-kappa B-inducing kinase (NIK), although later evidence has cast doubt on this. More recent reports show that mutations in IKAP such as a frameshift leading to a truncated protein or a missense mutation that leads to defective phosphorylation are responsible for the autosomal recessive genetic disease familial dysautonomia (FD). Reports indicating that it forms part of the RNA polymerase II transcription elongation complex suggest that this disease may be due to compromised transcription elongation. More recently, it was shown that IKAP associates with c-Jun N-terminal kinase (JNK) and could specifically enhance JNK activation induced by the upstream JNK activators MEKK1 and ASK1, indicating another possible cause for FD. At least two isoforms of IKAP are known two exist.

CCR5 Antibody: Human immunodeficiency virus (HIV) and related virus require coreceptors, in addition to CD4, to infect target cells. Some G protein-coupled receptors including CCR5, CXCR4, CCR3, CCR2b and CCR8 in the chemokine receptor family, and four new human molecules GPR15, STRL33, GPR1 and V28 were recently identified as HIV coreceptors. Among them, CCR5 (CC-CKR-5) is a principal coreceptor for macrophage- and dual-tropic HIV-1 strains fusion and entry of human white blood cells. CCR5 is required for the infection by HIV-1, HIV-2, and SIV. The beta-chemokines RANTES, MIP-alpha and MIP-beta are the ligands for CCR5 and prevent infection by M-tropic HIV-1. CXC5 associates with the surface CD4-gp120 of HIV complex and leads to membrane fusion and virus entry of target cells. The amino-terminal domain and the extracellular loops of CCR5 serve as HIV biding sites. CCR5 messenger RNA is expressed in lymphoid organs and monocytes.

Macrophage inflammatory protein-1 beta (MIP-1 beta), also known as CCL4, is a member of the CC family of chemokines and is most closely related to CCL3 (MIP-1 alpha). Cellular sources of MIP-1 beta include activated leukocytes (monocytes and T and B cells), brain endothelial cells, and smooth muscle cells (Lukacs et al.; Menten et al.). MIP-1 beta, MIP-1 alpha, and RANTES have been shown to be major HIV-suppressive factors, possibly through the interactions of these chemokines with the receptor CCR5 on CD4+ T cells, which is also a major receptor for HIV entry into CD4+ T cells (Cocchi et al.; Menten et al.). MIP-1 beta attracts a variety of immune cells to sites of microbial infection. In addition to its chemotactic functions, MIP-1 beta induces the release of proinflammatory cytokines, mast cell degranulation, and NK cell activation (Schall et al.). In mice, recruitment of regulatory T cells to B cells and antigen-presenting cells by MIP-1 beta plays a central role in the initiation of T cell and humoral responses, and the depletion of regulatory T cells or MIP-1 beta results in deregulated humoral responses and production of autoantibodies (Bystry et al.).
