AttoPhos® AP Fluorescent Substrate System, utilising a highly sensitive fluorescent alkaline phosphatase (AP) substrate, maintains a low fluorescence signal until enzymatically acted upon, yielding detection of AP to as little as 0,1 attomole.
- Excitation is at 435 nm, emission at 555 nm and large stokes shift (~120 nm)
- Linear kinetics over five orders of magnitude of alkaline phosphatase concentration
- Minimal signal fluorescence from interfering biological molecules
AttoPhos® Substrate: 2'-[2-benzothiazoyl]-6'-hydroxybenzothiazole phosphate [BBTP]) is cleaved by alkaline phosphatase to produce inorganic phosphate and the alcohol, 2'-[2- benzothiazoyl]-6'- hydroxybenzothiazole (BBT). This enzyme catalysed conversion is accompanied by flourescence properties that includes excitation and emission.
AttoPhos® Buffer: Buffer is optimised for sensitive detection of alkaline phosphatase when used to prepare working solutions with substrate. It contains 2,4 M DEA, 0,057 mM MgCl2, 0,005% NaN3 (pH 10).
