The P450-Glo CYP2C19 screening system provides a complete set of reagents for performing luminescent cytochrome P450 assays. The membranes are prepared from baculovirus-infected insect cells and contain human cytochrome P450, P450 reductase, and cytochrome b5. It also contains a membrane fraction devoid of cytochrome P450 activity as a negative control. The assays are ideal for testing the effects of drugs and new chemical entities on cytochrome P450 enzyme activities.
Delivery: Assay system contains Luciferin-H EGE (1×123 or 2×123 µg); luciferin detection reagent (lyophilised) (1 vial); reconstitution buffer with esterase (1×10 or 1×50 ml). Screening system contains P-Glo CYP2C19 assay includes Luciferin-H EGE (2×123 µg); luciferin detection reagent (lyophilised) (1 vial); reconstitution buffer with esterase (50 ml) and human CYP2C19 enzyme system includes CYP2C19 (1 pmol/µl), reductase and b5 (250 µl); control membranes (100 µl); potassium phosphate buffer, 1M (pH 7.4) (5 ml); luciferin-free water (50 ml); NADPH regeneration system solution A (2,75 ml); NADPH regeneration system solution B (0,6 ml).
