AccuStart™ II GelTrack PCR SuperMix is a 2X concentrated, ready-to-use reaction cocktail for routine PCR amplification of DNA fragments up to 4 kb followed by analysis on agarose gels. It contains all components, except primers and template. AccuStart™ II GelTrack PCR SuperMix simplifies reaction assembly, improves assay reproducibility, and reduces the risk of contamination. The supermix includes electrophoresis tracking dyes that migrate at approximately 4 kb and 50 bp to allow direct loading of PCR product on agarose gels following amplification. AccuStart™ II Taq DNA polymerase in the master mix is inactivated with monoclonal antibodies that bind the polymerase and keep it inactive prior to the initial PCR denaturation step. Upon heat activation (1 minute at 94 °C), the antibodies denature irreversibly, releasing fully active, unmodified Taq DNA polymerase. This enables specific and efficient primer extension with the convenience of room temperature reaction assembly.

Plasmid isolated with traditional purification procedures normally contain high levels of endotoxins (also known as lipopolysaccharides or LPS) that can significantly interfere with transfection experiments downstream. The E.Z.N.A.® Endo-Free plasmid mini kit II integrates an efficient endotoxin removal step into the plasmid purification procedure to produce high-quality transfection grade (<0,1 EU/µg) plasmid for efficient transfection. The bacterial cells are lysed using the alkaline-SDS lysis method. The cleared cell lysate is then treated with ETR reagent to efficiently remove the endotoxins. After adjusting the binding condition, the cell lysate is applied into the HiBind® DNA column and purified DNA is eluted from the column membrane.

The CloneJET PCR cloning kit contains a ready-to-use positive selection cloning vector pJET1.2/blunt. The vector contains a lethal restriction enzyme gene that is disrupted by ligation of a DNA insert into the cloning site. As a result, only bacterial cells with recombinant plasmids are able to form colonies. Recircularised pJET1.2/blunt vector molecules lacking an insert express a lethal restriction enzyme, which kills the host E. coli cell after transformation. This positive selection drastically accelerates the process of colony screening and eliminates additional costs required for blue/white selection.
