Mouse NGF (2.5S) was isolated from mouse submaxillary glands by method of Mobley et al (1976) and is a form of beta-NGF that has identical biological properties. NGF is known to regulate the survival and development of certain sympathetic and sensory neurons. It is a dimer with 2 identical polypeptide chains and dimeric molecular weight of approximately 26,500 Da. Isolation and purification of NGF from mouse submaxillary glands yields preparations of NGF (2.5S) with identical biological activity but with cleavages at the amino terminus (with the loss of 8 amino acids) and/or at the carboxy-terminus (with the loss of arginine). Microcystins are a group of cyclic heptapeptide hepatotoxins produced by a number of cyanobacterial genera. The most notable of which, and namesake, is the widespread genus Microcystis. Structurally, all microcystins consist of the generalized structure cyclo(-D-Ala1-X2-D-MeAsp3-Y4-Adda5-D-Glu6-Mdha7-). X and Y are variable L-amino acids, D-MeAsp is D-erythro-β-methylaspartic acid and Mdha is N-methyldehydroalanine. Adda is the cyanobacteria unique C20 β-amino acid 3-amino-9-methoxy-2,6,8-trimethyl-10-phenyl-deca-4,6-dienoic acid. Substitutions of the variable L-amino acids at positions 2 and 4 give rise to at least 21 known primary microcystin analogs and alterations in the other constituent amino acids result in more than 90 reported mycrocystins to date.

Fluor de Lys® Substrate is a highly sensitive and convenient alternative to radiolabeled, acetylated histones or peptide/HPLC methods for the assay of histone deacetylases. The Fluor de Lys® Substrate, which comprises an acetylated lysine side chain, is incubated with a sample containing HDAC activity (nuclear or cellular extract, purified enzyme, bead-bound immunocomplex, etc.). Deacetylation of the substrate sensitizes the substrate so that, in a second step, treatment with the Fluor de Lys® Developer produces a fluorophore. Histone deacetylases from all known classes have been successfully assayed with the Fluor de Lys® Substrate. These include human HDACs 1, 2, 3 and 8 (class I), human HDACs 4 and 9 (class II) and the sirtuins yeast Sir2, human SIRT1 and human SIRT2 (class III; reaction must include the sirtuin substrate NAD+). The substrate is cell-permeable and serves as the basis for the HDAC Cellular Activity Assay Kit (Prod. No. BML-AK503). NOTE: Must be used in conjunction with Fluor de Lys® Developer (Prod. No. BML-KI105). Sufficient for 100-1000 assays of HDACs or sirtuins (50 µl reaction volume, 50-500 µM (substrate).

Cytokines for expansion and cell culture of human hematopoietic stem cells. Hematopoietic stem cells are self renewing pluripotent cells often characterised by the expression of CD34. Studies have shown that decreased expression of CD34 tends to correlate with decreased self renewal properties and pluripotent potential. Culturing CD34+ cells in a cytokine mixture of TPO, SCF, and FLT-3 Ligand for short time periods allows CD34+ cells to proliferate without decreasing CD34 expression, thus retaining their stem cell properties. The combination of these cytokines allows for the ex vivo expansion of the most immature hematopoietic stem cells in cord blood or bone marrow preparations, but can be supplemented with other cytokines to promote specific lineages. For example, GM-CSF and/or G-CSF can be added to promote monocytes and granulocytes while EPO can be added to promote erthroid cells.

Neutrophil gelatinase-associated lipocalin(LCN2) is a secreted protein and belongs to the calycin superfamily. This protein is released from injured tubular cells after various damaging stimuli, is already known by nephrologists as one of the most promising biomarkers of incoming Acute Kidney Injury (AKI). Recent evidence also suggests its role as a biomarker in a variety of other renal and non-renal conditions. Moreover, recent studies seem to suggest a potential involvement of this factor also in the genesis and progression of chronic kidney diseases. NGAL is the first known mammalian protein which specifically binds organic molecules called siderophores, which are high-affinity iron chelators. NGAL, first known as an antibacterial factor of natural immunity, and an acute phase protein, is currently one of the most interesting and enigmatic proteins involved in the process of tumor development. acting as an intracellular iron carrier and protecting MMP9 from proteolytic degradation, NGAL has a clear pro-tumoral effect, as has already been observed in different tumors (e.g. breast, stomach, oesophagus, brain) in humans. In thyroid carcinomas, NGAL is strongly induced by NF-kB, an important factor involved both in tumor growth and in the link between chronic inflammation and neoplastic development. Thus, Lipocalin-2 (LCN2/NGAL) has been implicated in a variety of processes including cell differentiation, proliferation, survival and morphogenesis.
