Caspases are a family of cysteine proteases that can be divided into the apoptotic and inflammatory caspase subfamilies. Unlike the apoptotic caspases, members of the inflammatory subfamily are generally not involved in cell death but are associated with the immune response to microbial pathogens. Members of this subfamily include caspase-1, -4, -5, and -12. Activation of these caspases results in the cleavage and activation of proinflammatory cytokines such as IL-1b and IL-18. Caspase-4 was initially identified as a homologous protein to Caspase-1 and the C. elegans Ced-3 which could induce apoptosis in transfected cells. More recent studies have shown that it can be activated by ER stress and has been suggested to be involved in multiple neuronal pathologies such as Alzheimer's disease.

Insulin-growth factor-like gene family is a new family of proteins consisting of four proteins in humans (IGFL1 to 4) and one in mice (mIGFL). mIGFL is expressed in normal skin in mice and further upregulated during inflammation responses in skin or after skin wounding. In human only IGFL1 expression is increased in psoriatic skin samples. mIGFL and human IGFL1 and 3 interact with specificity and high affinity to a novel receptor named IGF-like family receptor 1 (formerly TMEM-149). Analysis of the amino acid sequence of IGFLR1 indicated that this receptor is likely a novel member of the TNF-R family. IGFLR1 is expressed most abundantly on mouse T cells, suggesting that mIGFL and IGFL1 produced in the skin may potentially exert regulatory functions on T cell responses.

Selectins are a family of calcium-dependent type 1 transmembrane proteins. Endothelial (E)-selectin is a heavily glycosylated transmembrane protein expressed by activated endothelial cells in microvascular linings. E-selectin, along with P-selectin and L-selectin, initiate recruitment of circulating leukocytes from blood to sites of inflammation in the vascular lining through interaction with specific cell surface-associated carbohydrate determinants. E-selectin consists of an N-terminal type 1 lectin domain, an EGF-like domain, 6 sushi (CCP/SCR) domains, a transmembrane sequence, and a short cytoplasmic domain. Recombinant Human E-selectin is a 58.6 kDa protein containing 535 amino acid residues, corresponding to the extracellular portion of the full length protein. Due to glycosylation, E-selectin migrates at an apparent molecular weight of approximately 65-85 kDa by SDS-PAGE analysis under reducing conditions.

Matrix metalloproteinases (MMPs) belong to a family of secreted or membrane-associated zinc endopeptidases capable of digesting extracellular matrix components. MMP-14 (MT1-MMP), membrane-type MMP, plays an important role in tumor invasion. MMP-14 is expressed on the surface of invasive tumor cells, in stromal cells of human colon, breast, and head and neck carcinomas. MMP-14 is secreted as zymogen with a prodomain, a catalytic domain, a hinge region, a hemopexin-like domain, and a transmembrane domain. It can activate pro-MMP-23 and degrade a variety of substrates, including fibrillar collagens I, II, III, fibronectin, vitronectin and laminin-1.

A truncated human MMP-14 with His-tag was expressed in E. coli. The Mr on SDS-PAGE is 31-kDa. Incubation with 1 mM APMA at 37°C for 2 hr will activate MMP-14. Its activity can be measured by FRET peptides
