Sodium chloride is a commonly used chemical which is found widely in nature. It is considered to be an essential nutrient. Excess amounts of sodium chloride can destroy electrolyte balance and cause death in most animals, including humans.
Sodium chloride is used in a wide variety of biochemical applications, including intravenous fluids (normal saline is 0.9% w/v in water 10), density gradients 3,6, a diluent to increase ionic strength in buffers or culture media and in salt-out procedures in the isolation of DNA. It has also been used in high concentrations for preservation of foods since bacteria cannot grow in high salt conditions. A salt-ice mixture in the ratio of approximately 33 g sodium chloride to 100 g ice (at -1°C) will drop in temperature to as low as -21°C, depending on the rate of stirring and the size of the ice chunks.

PMA (Phorbol-12-Myristate-13-Acetate) is a diester of phorbol and is a tumor promoting compound extracted from croton oil. It is a reversible, highly potent protein kinase C (PKC) activator in vitro and in vivo at nM concentrations. PMA’s effects on PKC are attributed to its similarity to diacylgylerol, a natural activator of PKC.
PMA activates Ca2+- ATPase and potentiates forskolin-induced cAMP formation. It has been shown to inhibit apoptosis induced by the Fas antigen, but PMA induces apoptosis in HL-60 promyelocytic leukemia cells.

DL-Dithiothreitol is also known as Clelands reagent; Protective agent for sulfhydryl groups (-SH). Quantitatively reduces disulfides (-S-S- to -SH). In this reaction the DTT is oxidized to the cyclic disulfide which ensures the reduction of other disulfides in solution. Disulfide reduction occurs quickly at pH 8.
Dithiothreitol is useful for stabilizing sulfhydryl containing enzymes. Effective in sample buffers for reducing protein disulfide bonds prior to SDS-PAGE. DTT can also be used for reducing the disulfide bridge of the cross-linker N,N'-bis(acryloyl)cystamine to break apart the matrix of a polyacrylamide gel. DTT is less pungent and is less toxic than 2-mercaptoethanol.
Useful for stabilizing sulfhydryl-containing enzymes.

Glycine is a non-essential amino acid. It is only amino acid with no asymmetric carbon and thus is not chiral. It is the major inhibitory neurotransmitter. It is involved in the biosynthesis of the porphyrin rings of hemes and chlorophylls.
Glycine is commonly used in buffer solutions, in electrophoresis and preparative chromatography. A study of the folding of monoclonal antibodies in the presence of glycine and their subsequent purification has been published. The use of glycine in the purification of lipopolysaccharides, lipooligosaccharides, and lipid A has been reported. It is an amino acid for use in cell culture media development applications and existing media formulations. Glycine is commonly used as a component in Tris-glycine and Tris-glycine-SDS running buffers for polyacrylamide gel electrophoresis, a component of Towbin's transfer buffer for Western blots, a buffer substance in cryoenzymology, in osmotic pressure maintenance in isoelectric focusing of erythrocytes, salting-in effect in protein chemistry, and as a buffer component in the coupled phosphatase-kinase reaction for end labelling of restriction fragments. Brefeldin A is a fungal metabolite which is a macrocyclic lactone exhibiting a wide range of antibiotic activity. Produced by Penicillium brefeldianum. Blocks binding of the cytosolic coat protein b-COP and ARF to Golgi membranes mediated by protein G. Also blocks protein transportation into post-Golgi compartments. It activates the sphingomyelin cycle. Brefeldin A mediated apoptosis has been observed in human tumor cells.
Brefeldin A reversibly inhibits the intracellular translocation of proteins in eukaryotes, e.g., during transport of proteins to the cell surface for secretion or expression. It has been reported to block the response of cultured cells to cholera toxin. In HepG2 cells, BFA induces two blocks in the secretory pathway; one at the level of the endoplasmic reticulum-Golgi juncture and the other in the trans-Golgi network. Trypan Blue is a blue acid dye with a strong affinity for cellulose containing substrates such as cotton; less affinity for proteinaceous materials. Trypan blue solution may be used in trypan blue based cytotoxicity and proliferation assays.
Trypan Blue is used as a vital dye which is especially important because it is taken up by the reticuloendothelial system. Clark describes assays for the study of teratogenic action of trypan blue on embryonic tissues using Davis and Sauter's fluorescence method and for the staining of collagen, including very fine fibrils, muscle and cornified epithelium using the Van Gieson method. Trypan blue is also recommended for use in dye exclusion procedures for viable cell counting. Non-viable cells will up-take trypan blue at a faster rate than viable cells.
