CD4 is a type I transmembrane glycoprotein expressed on most thymocytes and the helper/inducer subpopulation of mature T lymphocytes of all mouse strains tested. CD4 has also been detected at low density on pluripotent hematopoietic stem cells, bone marrow myeloid precursors, and intrathymic lymphoid precursors. The CD4 antigen functions as a co-receptor, interacting with class II major histocompatability complex (MHC) molecules in the recognition of foreign antigens by T cells. In vivo and in vitro treatment with the GK1.5 monoclonal antibody effectively depletes CD4+ cells. GK1.5 also blocks helper T cell responses to MHC class II antigens, including cytolysis, proliferation, allogeneic B cell help, and release of lymphokines.

PY20 binding to phosphorylated tyrosines can be inhibited by free phosphotyrosine and phenylphosphate but not by phosphoserine, phosphothreonine, or free phosphate.

CD5, also known as Lyt-1, is a monomeric type I transmembrane glycoprotein expressed on thymocytes, T lymphocytes, and a subset of B lymphocytes, but not on natural killer (NK) cells. It has been identified as the major ligand of the B-cell antigen CD72. The frequency of CD5+ B cells exhibits strain-dependent variation, and the phenotypic, anatomical, functional, developmental, and pathological characteristics of the CD5+ B cells suggest that they may represent a distinct lineage, known as B-1 cells. Binding of CD5 on the T cell surface can augment alloantigen- or mitogen-induced lymphocyte proliferation and induces increased cytosolic free calcium, IL-2 secretion, and IL-2R expression. It has been proposed that CD5 negatively regulates signal transduction mediated by the T-cell and B-cell receptors.

Chicken CD28 antigen is a type I transmembrane monomeric glycoprotein which does not contain the cysteine residues that form disulfide-linked covalent homodimers found in the mammalian counterpart. This may reduce the avidity of binding to individual ligand molecules expected to be present on antigen presenting cells. Peripheral γδ+ T cells are CD28 negative. Monoclonal antibodies against CD28 have a costimulatory effect on T cells stimulated by phorbol myristate acetate (PMA), concanavalin A, or monoclonal antibodies against the avian T cell receptor (TCR). Both Vβ1- and Vβ2-expressing cells respond equally well to stimulation with anti-CD28 in combination with PMA. These responses are resistant to cyclosporin A but inhibited by herbimycin A suggesting that CD28 employs a signaling pathway at least partly distinct from that triggered by TCR/CD3. The monoclonal antibody AV7 also reacts with turkey CD28 as demonstrated by flow cytometry.

Chicken Bu-1a, a product of the Bu-1a allele, is expressed on the cell surface as a disulfide-linked homodimer. In 10-week-old H.B15 and H.B2 chickens, Bu-1 is found on 85-90% of bursal cells, 2-8% of thymocytes, 15-27% of spleen cells, and 2-18% of peripheral blood cells. It is also expressed on subsets of macrophages and monocytes but not on granulocytes, erythrocytes, or thrombocytes. In chickens heterozygous for the Bu-1 alleles (Bu-1a/b), Bu-1a does not exhibit allelic exclusion. The monoclonal antibody 21-1A4 does not react with cells from CHA and H.B14ab strains by immunofluorescence.

Chicken CD45 is a transmembrane glycoprotein expressed on all leukocytes. B lineage cells in the bursa of Fabricius express the 215 kDa isoform while T lineage cells bear the 190 kDa variant. These high molecular weight molecules have intrinsic phosphotyrosine phosphatase activity characteristic of mammalian CD45. Levels of CD45 expression detected by the monoclonal antibody LT40 in the avian thymus are heterogeneous with approximately 90% of thymocytes expressing fourfold higher levels of surface CD45 (CD45hi) than do the remaining 10% (CD45low) of thymocytes. The CD45low population contains exclusively thymocytes with the CD3-CD4-CD8low phenotype characteristic of the immediate precursors to the CD3-CD4+CD8+ thymic population which are CD45hi. This shift from low to high levels of surface CD45 expression is concomitant with the transition from CD4-CD8low to CD4+CD8+ and before the expression of CD3. The ability of the monoclonal antibody LT40 to discriminate this CD45low subpopulation provides a suitable marker for the CD3-CD4-CD8low immediate precursors to the CD3-CD4+CD8+ thymocytes.

CD22, also known as Lyb-8, is a heterodimeric transmembrane glycoprotein and a member of a structurally related group of immunoglobulin (Ig) superfamily domain-containing proteins called the sialoadhesin family. It is detected in the cytoplasm early in B cell development (late pro-B cell stage), appears on the cell surface concomitant with IgD, and is found on most IgM+IgD+ mature B lymphocytes. Expression is lost with terminal differentiation of B cells and is absent on plasma cells. Activation of B cells via cross-linking of surface Ig increases CD22 expression. CD22 associates with the B cell receptor (BCR) complex and mediates intercellular adhesion. Its intracellular domain is phosphorylated after antigen receptor cross-linking and is involved in negative regulation of B cell activation.
