The monoclonal antibody TCR-1 precipitates a heterodimer of Mr 90 kDa (two bands of Mr 50 kDa and 40 kDa upon reduction) on chicken peripheral blood T cells. Deglycosylation of the heterodimer yields two polypeptides of Mr 35 kDa and 32 kDa. Unlike human and mouse in which γδ cells comprise a minor subset of T lymphocytes in the circulation, the chicken has a relatively large subset of γδ T cells. The frequency of TCRγδ cells is typically 20-25% of the total blood T cells but may reach approximately 50% in chickens of six- months of age. The majority of TCRγδ+ cells in the thymus and blood are CD4-CD8- although a small subset of them may express CD8 or CD4 coreceptors. However, when the TCRγδ+ cells migrate into the spleen and intestine most of them begin to express CD8. Although the biological function of γδ T cells is unclear, they are clearly capable of cytotoxic activity in vitro. The CD8+ γδ T cells may also be involved in down-regulation of the immune response. They cannot, however, induce a graft-vs-host (GVH) reactions.

Chicken CD45 is a transmembrane glycoprotein expressed on all leukocytes. B lineage cells in the bursa of Fabricius express the 215 kDa isoform while T lineage cells bear the 190 kDa variant. These high molecular weight molecules have intrinsic phosphotyrosine phosphatase activity characteristic of mammalian CD45. Levels of CD45 expression detected by the monoclonal antibody LT40 in the avian thymus are heterogeneous with approximately 90% of thymocytes expressing fourfold higher levels of surface CD45 (CD45hi) than do the remaining 10% (CD45low) of thymocytes. The CD45low population contains exclusively thymocytes with the CD3-CD4-CD8low phenotype characteristic of the immediate precursors to the CD3-CD4+CD8+ thymic population which are CD45hi. This shift from low to high levels of surface CD45 expression is concomitant with the transition from CD4-CD8low to CD4+CD8+ and before the expression of CD3. The ability of the monoclonal antibody LT40 to discriminate this CD45low subpopulation provides a suitable marker for the CD3-CD4-CD8low immediate precursors to the CD3-CD4+CD8+ thymocytes.

CD11a, also known as leukocyte function-associated antigen-1 (LFA-1α), represents the 180 kDa integrin αL subunit which combines with the CD18 β2 integrin subunit to form the αLβ2 integrin heterodimer. It is expressed on lymphocytes, granulocytes, monocytes, and macrophages and is upregulated on activated T cells. CD11a mediates adhesion of lymphocytes to vascular endothelium and is involved in costimulation.

CD3, a member of the immunoglobulin superfamily of cell surface receptors, is comprised of five invariable chains ranging from 16-28 kDa and is closely associated with the T cell antigen receptor (TCR). CD3 is expressed on 70-80% of normal peripheral blood lymphocytes and on 10-20% of thymocytes. It plays a major role in signaling during antigen recognition, leading to T cell activation.
