CD49f is the 125 kDa integrin α6 chain which associates noncovalently with the integrin β1 subunit (CD29) and with the integrin β4 (CD104) chain to form, respectively, the very late antigen-6 (VLA-6) and α6β4 integrin heterodimers. VLA-6 is expressed on thymocytes, T lymphocytes, monocytes, and platelets. CD49f/CD29 expression is upregulated on activated T lymphocytes and memory T cells. VLA-6 serves as the laminin receptor on platelets, monocytes, and T cells. VLA-6-mediated binding to laminin provides a costimulatory signal to T cells for activation and proliferation.

The monoclonal antibody BB6-10A10 stains the following percentages of surface immunoglobulin (sIg) positive cells in porcine lymphoid tissues - 11-15% in peripheral blood, 22-23% in mesenteric lymph nodes, 79-87% in ileal Peyer’s patches (ILPP), 6-17 in spleen, and 0% in thymus. While identification of the antigen(s) that are found predominantly on ILPP B cells and which are recognized by BB6-10A10 is speculative, the antibody appears to react with an immature population of B cells in the follicles of ILPPs and with a subpopulation of sIglow B cells in lymph nodes.

ApoScreen® Annexin V (SB Cat. No. 10040) is a recombinant protein that is expressed in E. coli. A number of changes in cell structure occur during apoptosis. One major change is the loss of membrane phospholipid asymmetry, with translocation of phosphatidylserine (PS) from the inner leaflet of the phospholipid bilayer to the external surface. Annexin V, a member of the annexin family of calcium-dependent phospholipid binding proteins, has a high affinity for phosphatidylserine (PS)-containing phospholipid bilayers. Conjugated Annexin V can be used to monitor changes in cell membrane phospholipid asymmetry, thereby providing a convenient tool for detection of apoptotic cells.

CD4 is a type I transmembrane glycoprotein expressed on most thymocytes and the helper/inducer subpopulation of mature T lymphocytes of all mouse strains tested. CD4 has also been detected at low density on pluripotent hematopoietic stem cells, bone marrow myeloid precursors, and intrathymic lymphoid precursors. The CD4 antigen functions as a co-receptor, interacting with class II major histocompatability complex (MHC) molecules in the recognition of foreign antigens by T cells. In vivo and in vitro treatment with the GK1.5 monoclonal antibody effectively depletes CD4+ cells. GK1.5 also blocks helper T cell responses to MHC class II antigens, including cytolysis, proliferation, allogeneic B cell help, and release of lymphokines.

CD19 is a monomeric transmembrane glycoprotein expressed at relatively constant levels throughout B cell development from early pro-B/pre-B cells (i.e. B220+/CD43+/HSA+) through fully differentiated B cell stages. Terminally differentiated plasma cells do not express CD19. In humans, the CD19 molecule on the surface of mature B cells associates with CD21 (CR-2) and CD81 (TAPA-1), and this multimolecular complex synergizes with surface immunoglobulin to provide signal transduction and promote cellular activation. All splenic and peritoneal IgM+ cells of both B-1 and B-2 lineages are CD19+, with B-1 cells expressing higher levels of CD19 than B-2 cells in these sites. Studies with CD19-deficient mice have suggested that this molecule may not be required for normal generation and maturation of B cells in the bone marrow. The monoclonal antibody 6D5 recognizes the same epitope as the published clone, 1D3.

CD24 is also present on mouse granulocytes, monocytes, Langerhans cells and erythrocytes. Its ligand is P-selectin (CD62P). CD24 may play a role in regulation of B cell proliferation and differentiation, and it has been proposed that heat stable antigen promotes homotypic adhesion between B lymphocytes.

The CD45 antigen is an essential regulator of leukocyte activation and development and is now known to be a protein tyrosine phosphatase, e.g., CD45 is able to dephosporylate the Src family of protein kinases. This activity is known to be required for signal transduction induced by T and B cell antigen receptor engagement, cytokine signaling, and Fc receptor stimulation. The functional state of CD45 also has a pronounced effect on lymphocyte development. CD45 exists in multiple forms, such as the restricted variants (CD45R; see companion products 1660, 1665, 1675, 1685 and 1820) and as allelic forms CD45.1 (Ly-5.1) and CD45.2 (Ly-5.2). The specificity of antibodies to CD45.1 and CD45.2 has been exploited in studies where resolution of donor and recipient cells is essential, such as in gene therapy or bone marrow transplantation. The monoclonal antibody 104 recognizes CD45 (leukocyte common antigen) on all leukocytes of mouse strains expressing the CD45.2 allotype (e.g., A, AKR, BALB/c, CBA/Ca, CBA/J, C3H/He, C57BL, C57BR, C57L, C58, DBA/1, DBA/2, NZB, SWR, 129).
