3,3',5,5'-Tetramethylbenzidine (TMB) ≥99%, Powder
Supplier: MP Biomedicals
3,3',5,5'-Tetramethylbenzidine is suitable as peroxidase substrate for use in ELISA procedures. The substrate produces a soluble end product that is pale blue in color and can be read spectrophotometrically at 370 or 620 to 650 nm. The TMB reaction may be stopped with 2 M H2SO4 (resulting in a yellow color), and read at 450 nm. A sensitive and specific reagent for the detection of blood, assay of hemoglobin, assay of peroxidases. It is useful in gel staining procedure for low levels of home-associated peroxidases and for enzyme immunoassay of horseradish peroxidase.

Phenol red sodium salt solution 0.5%, dark red solution
Supplier: MP Biomedicals
Phenol red is an acid-base indicator. It is made by condensing two moles of phenol with one mole of o-sulfobenzoic acid anhydride.
Phenol Red is used as a pH indicator in cell culture applications. A solution of phenol red will have a yellow color at a pH of 6.4 or below and a red color at a pH of 8.2 and above. Phenol red in tissue culture media can act as a weak estrogen, especially with human breast cancer cells. Phenol red can be used to measure hydrogen peroxide in cultured macrophages in multiwell plates.

Sodium pyruvate ≥99%, white powder cell culture reagent
Supplier: MP Biomedicals

Pyruvic acid is an intermediate in sugar metabolism and in enzymatic carbohydrate degradation (alcoholic fermentation) where it is converted to acetaldehyde and CO2 by carboxylase. In muscle, pyruvic acid (derived from glycogen) is reduced to lactic acid during exertion, which is reoxidized and partially retransformed to glycogen during rest. It improves coliform recovery when present in culture medium. It is involved in a metabolic regulatory pathway activated by mitochondrial oxidants. Pyruvate is involved in respiratory regulation in plants by interacting with alternative oxidase at a conserved cysteine residue. It may help prevent hydrogen peroxide mediated cell death.
Sodium pyruvate is utilized as a component in culture broth and media. Sodium pyruvate is used in Wallen fermentation medium to enhance the conversion of oleic acid to 10-ketostearic acid by Bacillus sphaericus. Sodium pyruvate has also been used to establish stably transfected human B cell lines.
Sodium Pyruvate has shown antioxidant properties and protective effects against oxygen radicals. Pyruvate is produced as part of glycolysis and is an intermediate in many metabolic pathways. It can be converted into acetyl CoA and enter the TCA Cycle.

Sodium selenite ∼98%, powder cell culture reagent
Supplier: MP Biomedicals
Sodium selenite is commonly used as a source of selenium in biological research, where selenium is an essential trace element that is normally provided by serum. Selenium is present in selenoproteins such as glutathione peroxidase and thioredoxin reductase, which contain the selenium analog of cysteine, selenocysteine. In particular, glutathione peroxidase has a role in detoxification in vivo as a scavenger of peroxides. Sodium selenite is included in various medium supplements for use in cell culture. Sodium selenite has been utilized in studies of cell proliferation and cancer research. It has been used to alter gene expression in HepG2 cells as analyzed by cDNA microarrays. Sodium selenite can inhibit zinc finger protein/DNA interactions. Sodium selenite has been shown to alter mitochondrial membrane potentials and thus potentially contribute to apoptosis.

Ferritin (equine from horse spleen, thick), dark brown liquid
Supplier: MP Biomedicals
Ferritin, Equine is a shell of 24 protein subunits (apoferritin) and a core Fe 3+ ions with a peptide MW of 440 kDa. Review of the ferritin-like superfamily of iron-storage proteins, virtually ubiquitous in life on earth.
A ubiquitous iron storage protein that plays a key role in iron metabolism. It serves as an intracellular iron reserve (particularly in spleen, liver, intestinal mucosa, and bone marrow) and functions in iron detoxification. Studies have shown that ferritin iron incorporation is mediated by a ferroxidase activity associated with ferritin H subunits and a nucleation center associated with ferritin L subunits. Release of iron from ferritin has an essential role in iron-dependent lipid peroxidation and may contribute to free radical-induced cell damage in vivo. Therefore, by binding iron, ferritin may function as an antioxidant.
