TIGAR Antibody: The p53 tumor-suppressor gene integrates numerous signals that control cell life and death; loss of its functions contributes to the development of most cancers. Recent studies have demonstrated the ability of p53 to regulate the expression of several proteins involved in glycolysis and oxidative phosphorylation, such as TIGAR, SCO2, and phosphoglycerate mutase. TIGAR is a recently discovered protein that functions to regulate glycolysis and protect cells against oxidative stress. TIGAR is similar in structure to proteins in the phosphoglycerate mutase family, most notably 6-phosphofructo-2-kinase, suggesting TIGAR may function as a fructose bisphosphatase. Expression of TIGAR in transfected cells correlated with an inhibition of glycolysis and decreased levels of reactive oxygen species and p53-induced apoptosis, indicating that TIGAR may act to modulate the apoptotic response to p53, thereby allowing cells to survive mild or transient stresses.

Syntaxin 13 is an integral membrane protein that belongs to the t-SNARE family, a group of proteins involved in protein transport. Confocal immunofluoresence and electron microscopy studies have shown that syntaxin 13 is primarily localized to tubular early and recycling endosomes, where it colocalizes with transferrin receptor, and it is also localized in endosomal vacuoles. Syntaxin 13 has been found to be expressed in all tissues, with higher levels of the protein found in brain, lung, spleen, thymus and testes. Immunoprecipitation studies show that syntaxin 13 complexes with beta-SNAP, VAMP2/3, and SNAP25. The binding of this complex to alpha-SNAP and NSF is terminated in the presence of ATP. These results suggest that syntaxin 13 is a SNARE protein which mediates the recycling protein flow through tubulo vesicular recycling endosomes.

ATG101 Antibody: Autophagy, the process of bulk degradation of cellular proteins through an autophagosomic-lysosomal pathway is important for normal growth control and may be defective in tumor cells. It is involved in the preservation of cellular nutrients under starvation conditions as well as the normal turnover of cytosolic components. This process is negatively regulated by TOR (Target of rapamycin) through phosphorylation of autophagy protein ATG1. ATG101 is a recently discovered protein that stabilizes ATG13, another autophagy protein that forms a complex with the mammalian homologs of ATG1, ULK1 and ULK2, and with FIP200. This complex is a target of TOR phosphorylation under normal conditions; inhibition of TOR by rapamycin or leucine deprivation leads to dephosphorylation of ATG13, ULK1 and ULK2, which then leads to autophagy. ATG101 also interacts with ULK1 and is essential for autophagy.
