The integrin alpha 4 (also known as CD49d and ITGA4) belongs to the integrin alpha chain family of proteins. Integrins are heterodimeric integral membrane proteins composed of an alpha and beta chains (reviewed in 1). Alpha 4 (α4) chain associates with either beta 1 (β1) or beta 7 (β7) chain. It has been demonstrated that the putative ligand-binding sites of both integrin α4β1 and α4β7 is located on the α4 chain. These ligands included Madcam, VCAM, and fibronectin (2-4). Madcam is known as the principal ligand for integrin a4b7. Recently it was also demonstrated that HIV-1 envelope can mimic Madcam by binding to and signaling though integrin α4β7, the gut mucosal homing receptor for peripheral T cells (5). This recombinant protein fragment includes the ligand and monoclonal antibody binding domains; this binding has been tested and verified in ELISA assays. This recombinant protein is recognized PSI antibodies Integrin 4α (IN1) (PSI 4827) and Integrin 4α (IN2) (PSI 4837), but not Integrin 4α (CT) (PSI 4783).

AG-490 is a potent inhibitor of the JAK-2 tyrosine kinase. In acute lymphoblastic leukemia (ALL) cells, which abundantly express JAK-2, AG-490 dose-dependently inhibited DNA synthesis, blocked cell growth and induced apoptosis. At 5 µM, AG-490 almost completely blocked the growth of all pre-B ALL cells but had no significant effect on the growth of mitogen-stimulated normal B or T cells, B-cell lymphoma or T-cell leukemia cells. AG-490 does not significantly inhibit other kinases such as Lck, Lyn, Btk, Syk and Src. It blocks interleukin-7-induced JAK kinase activity in T-cells (JAK-1, JAK-3) and the consequent phosphorylation of PI-3 kinase. AG-490 is cell permeable and is a valuable tool for studying the cellular role of JAK kinases in signal transduction.

MyD88 is a general adapter protein for the Toll/IL-1R family of receptors and plays an important role in the inflammatory response induced by cytokines IL-1 and IL-18 and endotoxin. NKG2D Ligand 2 (N2DL2) is a member of a family of cell-surface proteins. N2DL2 function as ligands for human cytomegalovirus glycoprotein UL16. N2DL2 is anchored to the membrane via a GPI-linkage. N2DL2 is bind to human NKG2D, an activating receptor expressed on NK cells, NKT cells, T cells. Engagement of NKG2D results in the activation of cytolytic activity and cytokine production by these effects cells. Interleukin-33 (IL-33) is a recently identified member of the IL-1 family of cytokines whose other members include IL-1, IL-1Ra and IL-18 (1,2). Its receptor has been shown to be ST2, an IL-1 receptor family member that also acts as a negative regulator of TLR-IL-1R signaling (1,3) and IL-1R accessory protein (IL-1RAcP) (4). Receptor binding of IL-33 activates NF-B and MAP kinases and induces the expression of TH2-associated cytokines such as IL-4, IL-5 and IL-6. Prolonged IL-33 treatment of mice led to the development of eosinophilia, splenomegaly, and severe pathological changes in mucosal organs such as lungs, esophagus and small intestine (2). Recent experiments have shown that IL-33 can also co-localize with heterochromatin and possesses transcriptional repressor activities, indicating that IL-33 may function as both a proinflammatory cytokine and an intracellular nuclear factor with transcriptional regulatory properties (5). This recombinant protein represents the cleaved and presumably activated form of IL-33, but has not been tested in any biological assays.
