Anti-GSK3A Mouse Monoclonal Antibody [clone: 6D3]
Supplier: Enzo Life Sciences
Glycogen Synthase Kinase 3β (GSK-3β) is a unique serine/threonine kinase that is inactivated by phosphorylation. In response to insulin binding, PKB/AKT phosphorylates GSK-3β on serine 9, which prevents GSK-3β from phosphorylating glycogen synthase. Unphosphorylated glycogen synthase is active and able to synthesize glycogen. GSK-3β is also unique in that it requires a substrate that has been phosphorylated by a distinct kinase before it can phosphorylate the substrate. This phosphate priming mechanism explains why phosphorylation of serine 9 inactivates GSK-3β. The phosphorylated serine binds to the GSK-3β priming phosphate position and prevents binding of alternative substrates. In addition to insulin signaling, GSK-3β participates in the Wnt signaling pathway, where it forms a complex with axin, beta-catenin and adenomatous polyposis coli (APC) protein. In the presence of Wnts, GSK-3β is unable to phosphorylate beta-catenin, which leads to stabilization of beta-catenin. The Wnt pathway inactivates GSK-3β via the proteins, Dishevelled and FRAT, which disrupt the interaction of GSK-3β with axin, beta-catenin, and APC. Clinically, there is considerable interest in GSK-3β inhibitors because they may mimic the effect of insulin or reduce the hyperphosphorylation of Tau that is observed in Alzheimer's Disease.

Anti-KRT6A Mouse Monoclonal Antibody [clone: LHK6]
Supplier: Prosci
This antibody recognizes a protein of 56kDa, identified as cytokeratin 6 (CK6) or Keratin 6. In humans, multiple isoforms of Cytokeratin 6 (6A-6F), encoded by several highly homologous genes, have distinct tissue expression patterns, and Cytokeratin 6A is the dominant form in epithelial tissue. The gene encoding human Cytokeratin 6A maps to chromosome 12q13, and mutations in this gene are linked to several inheritable hair and skin pathologies. Keratins 6 and 16 are expressed in keratinocytes, which are undergoing rapid turnover in the suprabasal region (also known as hyper-proliferation-related keratins). Cytokeratin 6 is found in hair follicles, suprabasal cells of a variety of internal stratified epithelia, in epidermis, in both normal and hyper-proliferative situations. Epidermal injury results in activation of keratinocytes, which express Cytokeratin 6 and 16. Keratin 6 is strongly expressed in about 75% of head and neck squamous cell carcinomas. Expression of Cytokeratin 6 is particularly associated with differentiation.

Anti-H2AFX Rabbit Polyclonal Antibody
Supplier: Rockland Immunochemical
Histone H2A.X (phospho S139) antibody is ideal for western blotting and ELISA. Histones play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA is wrapped around histone-groups, consisting of the core histones H2A, H2B, H3 and H4. As a reaction on DNA Double-strand breaks (DSB) H2AX becomes phosphorylated on serine 139, called gamma-H2AX. ATM, ATR and PRKDCs, kinases of the PI3-family, are responsible for this phosphorylation. The modification can happen accidentally during replication fork collapse, exogenous genotoxic agents, may also occur during meiotic recombination events and immunoglobulin class switching in lymphocytes, in the response to ionizing radiation but also during controlled physiological processes such as V(D)J recombination. Mutagenesis experiments have shown that the modification is necessary for the proper formation of ionizing radiation induced foci in response to double strand breaks, but is not required for the recruitment of proteins to the site of DSBs. Gamma-H2AX is a sensitive target for looking at DSBs in cells. Dephosphorylation of Ser-140 by PP2A is required for DNA DSB repair. The role of the phosphorylated form of the histone in DNA repair is under. Anti-H2AX pS139 is ideal for researched interested in Histones, DNA Damage and Repair, and Epigenetics.

Supplier: Prosci
Twenty human keratins are resolved with two-dimensional gel electrophoresis into acidic (pI 6.0) subfamilies. This antibody detects acidic (Type I or LMW) and basic (Type II or HMW) cytokeratins: 67kDa (CK1); 64kDa (CK3); 59kDa (CK4); 58kDa (CK5); 56kDa (CK6); 52kDa (CK8); 56.5kDa (CK10); 50kDa (CK14); 50kDa (CK15); 48kDa (CK16); 40kDa (CK19). Many studies have shown the usefulness of keratins as markers in cancer research and tumor diagnosis. It is a broad spectrum anti pan-cytokeratin antibody, which differentiates epithelial tumors from non-epithelial tumors e.g. squamous vs. adenocarcinoma of the lung, liver carcinoma, breast cancer, and esophageal cancer. It may be useful to characterize the source of various neoplasms and to study the distribution of cytokeratin containing cells in epithelia during normal development and during the development of epithelial neoplasms. This antibody stains cytokeratins present in normal and abnormal human tissues and has high sensitivity in the recognition of epithelial cells and carcinomas.

Tricine ≥99%, white powder for electrophoresis
Supplier: MP Biomedicals
Tricine was first prepared by Good for use as a buffer for chloroplast reactions. It is structurally similar to Tris, but is much less inhibitory at high concentrations. For ATP assays using firefly luciferase, tricine buffer at 25 mM was found to be the best of ten common buffers tested.
Tricine is used as buffer component for separation of low molecular weight peptides. Tricine can be used in cryopreservation medium for the preservation of tissues and organs. Cryopreservation depends on the physical and chemical characteristics of the preservation medium used. The pH values and pK values for tricine/DMSO mixtures has been reported down to -20 °C. Tricine has been found to be an efficient scavenger of hydroxyl radicals in a study of radiation-induced membrane damage. Tricine is typically the buffer of choice in SDS-PAGE systems when separating proteins in the range of 1 to 100 kDa.
A buffer may be prepared by titrating with sodium hydroxide to the desired pH, using about a half-equivalent of NaOH.

Anti-BRCA1 Mouse Monoclonal Antibody [clone: MS13]
Supplier: Genetex
BRCA1 (breast and ovarian cancer susceptibility protein 1) is a RING finger protein containing a BRCT domain. BRCA1 exists as a heterodimer with 22 possible isoforms. The full length protein has a reported molecular weight of 208 kD. BRCA1 localizes to the mitotic spindle microtubules, centriole walls, pericentriolar fibers at centrosomes. Unphosphorylated BRCA1 localizes on chromosomes from metaphase through telophase; phosphorylated BRCA1 resides in inner chromosomal structure, centrosome, cleavage furrow during prophase through telophase, and relocalizes to the perinuclear region when cells are subjected to IR or UV radiation in S phase. BRCA1 acts as a tumor suppressor and can function as a secreted growth inhibitory protein, participate in transcription coupled repair of oxidative DNA damage, X-chromosome inactivation, and can function as a E3 ubiquitin ligase. BRCA1 can be transcriptionally downregulated by Ets-2, Brg-1, and Hmga-1. BRCA1 can be modified by glycosylation, ubiquitination and phosphorylation by CDK4, ATM/ATR, cdk2, and hChk2. The BRCA1 protein has been reported to interact with RNA polymerase II holoenzyme and BARD1. BRCA1 contains at least two nuclear localization signals and is proposed to be a tumor suppressor protein. It is a serine phosphoprotein that undergoes hyperphosphorylation during late G1 and S phases of the cell cycle and is transiently dephosphorylated early after M phase. BRCA1 protein alters in a qualitative and quantitative manner during cell cycle progression. The amount of BRCA1 protein is highest during S phase and remains elevated toward G2 / M, before it declines in early G1 phase. Inherited loss of BRCA1 function confers an increased susceptibility for both breast and ovarian cancer.

Folic acid ≥95%, orange powder
Supplier: MP Biomedicals
Folic acid, also known as folate, is a B vitamin that can be found in a variety of fruits and vegetables. It can also be chemically synthesized. Folate, a watersoluble vitamin, helps the body form red blood cells and aids in the formation of genetic material within every body cell. This product exhibits metal binding properties. Hematopoietic vitamin present, free or combined with one or more additional molecules of L-(+)-glutamic acid, in liver, kidney, mushrooms, spinach, yeast, green leaves, and grasses.
Folic acid (Vitamin B9 and folate) is essential to numerous bodily functions. The human body needs folate to synthesize DNA, repair DNA, and methylate DNA as well as to act as a cofactor in certain biological reactions. It is especially important in aiding rapid cell division and growth, such as in infancy and pregnancy.
Folic acid (FA) and dihydrofolic acid (FAH2) are substrates of dihydrofolate reductase(s) which reduce them to tetrahydrofolate (THF), which in turn supports ‘one carbon’ transfer. Tetrahydrofolates are required for de novo synthesis of purines, thymidylic acid and various amino acids and for post-translational methylation (epigenetics).
Very slightly soluble in cold water (0.0016 mg/mL at 25 °C), soluble to about 1% in boiling water. Slightly soluble in methanol, appreciably less soluble in ethanol and butanol. Insoluble in acetone, chloroform, ether, benzene. Relatively soluble in acetic acid, phenol, pyridine, solutions of alkali hydroxides and carbonates. Soluble in hot dilute HCl and H2SO4.
