The LASS (longevity assurance homolog) family members represent a subgroup of the homeobox gene family and are highly conserved from yeasts to mammals. Six members of this family of proteins have been characterized (LASS1-6) and all are involved in ceramide synthesis during cell growth regulation and cancer differentiation. Like the highly homologous LASS5, LASS6 is also an endoplasmic reticulum, multi-pass membrane protein. LASS6 is also involved in the synthesis of C14, C16 and C18-ceramide, but shows a preference for unsaturated fatty acids. LASS6 is broadly expressed in a wide range of tissues and microarray data suggests that it may play a role in cancer differentiation and early embryonic development.

XBP-1 Antibody: X box binding protein 1 (XBP-1) is a key protein in the mammalian unfolded protein response (UPR) that protects the cell against the stress of malfolded proteins in the endoplasmic reticulum (ER). Upon sensing unfolded proteins, an ER transmembrane endonuclease and kinase termed IRE1p is activated and excises an intron from XBP-1 mRNA. The spliced XBP-1 mRNA results in a 371 amino acid protein (XBP-1s) which is then translocated to the nucleus where it binds to the regulatory elements of downstream genes. Together with other UPR transcription factors such as ATF6, XBP-1 stimulates the production of ER stress proteins including the ER resident protein chaperones glucose regulated protein (GRP) 78 and GRP94.

APOBEC3G Antibody: The Apoliprotein B mRNA-editing, enzyme-catalytic, polypeptide-like (APOBEC) 3 is a multi-isoform member of the cytidine deaminase family of enzymes that act on monomeric nucleoside and nucleotide substrates. Similar to TRIM5alpha which targets incoming retroviral capsids, APOBEC3 plays a major role in cellular defense against retroviral infection as at least two isoforms, APOBEC3G and to a lesser extent APOBEC3F, can be incorporated HIV-1 virions and induce hypermutation in the newly synthesized viral DNA and thus destabilize the viral genome. This innate mechanism of retroviral resistance is counteracted by the HIV-1 Vif protein by inducing the ubiquitization and degradation of APOBEC3G; a single amino acid substitution (D128K) blocks APOBEC3G depletion without affecting its inhibitory activity.

FAM84A is a 292 amino acid protein that belongs to the FAM84 family of proteins. Predominantly expressed in testis, FAM84A shares 44% amino acid identity with the related protein FAM84B. FAM84A localizes to a subcellular membrane region where there is no contact between neighboring cells and is believed to play a role in cell morphology and motility. More specifically, the expression of FAM84A increases cell motility. Two FAM84A isoforms are expressed due to alternative splicing events. Isoform 2 can be phosphorylated on various serine residues and this phosphorylation is associated with cellular morphology. FAM84A is upregulated in colorectal cancer, lung cancer, pancreatic cancer, cholangiocarcinoma and bladder cancer tissues. Via its ability to increase cell motility, FAM84A may contribute to the invasion and metastasis of cancer cells.
