The LASS (longevity assurance homolog) family members represent a subgroup of the homeobox gene family and are highly conserved from yeasts to mammals. Six members of this family of proteins have been characterized (LASS1-6) and all are involved in ceramide synthesis during cell growth regulation and cancer differentiation. Like the highly homologous LASS5, LASS6 is also an endoplasmic reticulum, multi-pass membrane protein. LASS6 is also involved in the synthesis of C14, C16 and C18-ceramide, but shows a preference for unsaturated fatty acids. LASS6 is broadly expressed in a wide range of tissues and microarray data suggests that it may play a role in cancer differentiation and early embryonic development.

XBP-1 Antibody: X box binding protein 1 (XBP-1) is a key protein in the mammalian unfolded protein response (UPR) that protects the cell against the stress of malfolded proteins in the endoplasmic reticulum (ER). Upon sensing unfolded proteins, an ER transmembrane endonuclease and kinase termed IRE1p is activated and excises an intron from XBP-1 mRNA. The spliced XBP-1 mRNA results in a 371 amino acid protein (XBP-1s) which is then translocated to the nucleus where it binds to the regulatory elements of downstream genes. Together with other UPR transcription factors such as ATF6, XBP-1 stimulates the production of ER stress proteins including the ER resident protein chaperones glucose regulated protein (GRP) 78 and GRP94.

APOBEC3G Antibody: The Apoliprotein B mRNA-editing, enzyme-catalytic, polypeptide-like (APOBEC) 3 is a multi-isoform member of the cytidine deaminase family of enzymes that act on monomeric nucleoside and nucleotide substrates. Similar to TRIM5alpha which targets incoming retroviral capsids, APOBEC3 plays a major role in cellular defense against retroviral infection as at least two isoforms, APOBEC3G and to a lesser extent APOBEC3F, can be incorporated HIV-1 virions and induce hypermutation in the newly synthesized viral DNA and thus destabilize the viral genome. This innate mechanism of retroviral resistance is counteracted by the HIV-1 Vif protein by inducing the ubiquitization and degradation of APOBEC3G; a single amino acid substitution (D128K) blocks APOBEC3G depletion without affecting its inhibitory activity.

Gamma-aminobutyric acid type A (GABAA) receptors mediate inhibitory neurotransmission in the mammalian central nervous system. The receptor exists as a pentameric ion channel composed by heteromeric combinations of alpha, beta, gamma, delta, epsilon, theta, or pi subunits. Only specific subunit combinations produce viable receptors, while others never translocate to the cell surface from the ER where they are synthesized, and subsequently degraded. The theta subunit forms a receptor in combination with alpha3 subunits in monoaminergic cell groups. These receptors, found especially in the septum, preoptic areas, hypothalamic nuclei, amygdala and thalamus, likely have unique pharmacological properties linked to their expression in this particular cell type and not cholinergic cell groups, and may play a role in opiate withdrawl symptoms.

FAM84A is a 292 amino acid protein that belongs to the FAM84 family of proteins. Predominantly expressed in testis, FAM84A shares 44% amino acid identity with the related protein FAM84B. FAM84A localizes to a subcellular membrane region where there is no contact between neighboring cells and is believed to play a role in cell morphology and motility. More specifically, the expression of FAM84A increases cell motility. Two FAM84A isoforms are expressed due to alternative splicing events. Isoform 2 can be phosphorylated on various serine residues and this phosphorylation is associated with cellular morphology. FAM84A is upregulated in colorectal cancer, lung cancer, pancreatic cancer, cholangiocarcinoma and bladder cancer tissues. Via its ability to increase cell motility, FAM84A may contribute to the invasion and metastasis of cancer cells.

NPDC-1 (Neural Proliferation Differentiation and Control-1) is expressed in neurons once they have stopped dividing and begun to differentiate. NPDC-1 is transported from the Golgi apparatus via vesicles before becoming internalized by endosomes at the cell membrane. NPDC-1 interacts with Cdk2, D-type cyclins, and the transcription factor E2F1. This interaction can lead to an increased replication time, and might have implications in final neural differentiation and apoptosis. NPDC-1 has been shown to colocalize with synaptic vesicle proteins: synaptophysin, synaptobrevin 2, and Rab3 GEP (Rab3 GTP/GDP exchange protein). One function of NPDC-1 is to regulate retinoic acid-mediated events by directly interacting with retinoid receptors. The amino acid sequence of NPDC-1 is highly conserved between mouse, rat, and human.
