The deubiquitinylating enzyme USP19 is induced in skeletal muscle atrophy in response to numerous catabolic stimuli, including cancer, fasting, diabetes, and glucocorticoids. Loss of USP19 interferes with the growth of L6 myoblasts through its modulation of the transcription of major myofibrillar proteins.

USP19 regulates cell growth, partly through indirect control of p27Kip1 degradation in the G1 phase of the cell cycle. Since a number of cancers are associated with enhanced degradation of p27Kip1, the overexpression of USP19 may also be a pathogenetic mechanism of the disordered cell growth in cancer and a potential novel therapeutic target for its treatment.

In addition USP19 has been shown to be a tail anchored DUB, localised to the ER, suggesting a role in the turnover of ERAD substrates and to be targeted by the unfolded protein response.

USP19 is expressed in a wide range of tissues and is localised to the cytoplasm and endoplasmic reticulum.

The proteasome is widely recognised as the central enzyme of non-lysosomal protein degradation. It is responsible for intracellular protein turnover and it is also critically involved in many regulatory processes and, in higher eukaryotes, in antigen processing. The 26S proteasome is the key enzyme of the ubiquitin/ATP-dependent pathway of protein degradation. The catalytic core of this unusually large (2000kDa, 450Å in length) complex is formed by the 20S proteasome, a barrel shaped structure shown by electron microscopy to comprise of four rings each containing seven subunits.  Based on sequence similarity, all fourteen 20S proteasomal subunit sequences may be classified into two groups, α and β, each group having distinct structural and functional roles. The α-subunits comprise the outer rings and the β-subunits the inner rings of the 20S proteasome. Observations of the eukaryotic proteasome and analysis of subunit sequences indicate that each ring contains seven different subunits (α7β7β7α7) with a member of each sub-family represented in each particle. Each subunit is located in a unique position within the α- or β-rings. Lmp2, Lmp7 and MECL are interferon gamma-inducible catalytic subunits of the 20S proteasome which may replace the constitutive catalytic subunits, delta, X and Z respectively, during proteasome biogenesis. Lmp2 and Lmp7 alter the cleavage site specificity of the 20S proteasome and are required for the efficient generation of T cell epitopes from a number of viral proteins and for optimal MHC class I cell surface expression.

Tumor necrosis factor alpha (TNF-α), also known as cachectin, is a 17.5 kDa 157 amino acid member of the TNF superfamily of cytokines that is a potent lymphoid factor with effects on a wide range of target cells. Active TNFalpha is produced in both soluble and membrane-anchored trimers by macrophages, NK cells, and T- and B-lymphocytes. The diffusible free radical gas nitric oxide (NO) affects a variety of physiological functions, and is a key regulator of the cardiovascular, nervous, and immune systems. NO is synthesized in many tissues from L-arginine, oxygen, and NADPH by three known isoforms of a heme-containing flavoprotein termed NO synthase (nNOS/NOS-I, iNOS/NOS-II, and eNOS/NOS-III). nNOS is a constitutively expressed neuronal NOS isoform that exits in its latent form until it is activated by the binding of calmodulin follo wing elevation of intracellular calcium levels. The C-terminus of nNOS contains a conserved serine residue, Ser1417, analogous to Ser1177 of the constitutively expressed endothelial NOS isoform (eNOS). Phosphorylation of Ser1417 is believed to regulate nNOS activation, particularly in glucose-sensing neurons, where inhibition of AMPK pathways by glucose and leptin serve to suppress nNOS activity, whereas activation of AMPK by insulin leads to nNOS activation.
