IL-36Ra/IL-1F5 is a highly and a specific antagonist of the IL-1 receptor-related protein 2-mediated response to IL-36alpha (IL-1F6), IL-36beta (IL-1F8) and IL-36gamma (IL-1F9). These cytokines bind to IL-36R (IL-1Rrp2) and IL-1RAcP, activating similar intracellular signals as IL-1. IL-36Ra inhibits the production of proinflammatory cytokines, including IL-12, IL-1beta, IL-6, TNF-alpha and IL-23 induced by IL-36 in BMDC and CD4 T cells. Skin and dendritic cells are target of the IL-36 interleukins leading to a Th1 response. Recently mutations that affect the levels and the activity of IL-36Ra have been found in patients with pustular psoriasis, leading to enhanced production of inflammatory cytokines (IL-8 in particular) by keratinocytes.

Oncostatin M (OSM) is a growth and differentiation factor that participates in the regulation of neurogenesis, osteogenesis and hematopoiesis. Produced by activated T cells, monocytes and Kaposi’s sarcoma cells, OSM can exert both stimulatory and inhibitory effects on cell proliferation. It stimulates the proliferation of fibroblasts, smooth muscle cells and Kaposi’s sarcoma cells, but inhibits the growth of some normal and tumor cell lines. It also promotes cytokine release (e.g. IL-6, GM-CSF and G-CSF) from endothelial cells, and enhances the expression of low-density lipoprotein receptors in hepatoma cells. OSM shares several structural and functional characteristics with LIF, IL-6, and CNTF. Human OSM is active on murine cells. Recombinant Human Oncostatin M is a 25.7 kDa protein, containing 227 amino acid residues (full length precursor).

It is upregulated in cisplatin-resistant human bladder tumors, suggesting that its expression may be important for cisplatin resistance and inhibition of apoptosis in cancer cells. At least two isoforms of Bfl-1 are known to exist.

Matrix metalloproteinases (MMPs) belong to a family of secreted or membrane-associated zinc endopeptidases capable of digesting extracellular matrix components. MMP-14 (MT1-MMP), membrane-type MMP, plays an important role in tumor invasion. MMP-14 is expressed on the surface of invasive tumor cells, in stromal cells of human colon, breast, and head and neck carcinomas. MMP-14 is secreted as zymogen with a prodomain, a catalytic domain, a hinge region, a hemopexin-like domain, and a transmembrane domain. It can activate pro-MMP-23 and degrade a variety of substrates, including fibrillar collagens I, II, III, fibronectin, vitronectin and laminin-1.

A truncated human MMP-14 with His-tag was expressed in E. coli. The Mr on SDS-PAGE is 31-kDa. Incubation with 1 mM APMA at 37°C for 2 hr will activate MMP-14. Its activity can be measured by FRET peptides
