TIGAR Antibody: The p53 tumor-suppressor gene integrates numerous signals that control cell life and death; loss of its functions contributes to the development of most cancers. Recent studies have demonstrated the ability of p53 to regulate the expression of several proteins involved in glycolysis and oxidative phosphorylation, such as TIGAR, SCO2, and phosphoglycerate mutase. TIGAR is a recently discovered protein that functions to regulate glycolysis and protect cells against oxidative stress. TIGAR is similar in structure to proteins in the phosphoglycerate mutase family, most notably 6-phosphofructo-2-kinase, suggesting TIGAR may function as a fructose bisphosphatase. Expression of TIGAR in transfected cells correlated with an inhibition of glycolysis and decreased levels of reactive oxygen species and p53-induced apoptosis, indicating that TIGAR may act to modulate the apoptotic response to p53, thereby allowing cells to survive mild or transient stresses.

Noxa Antibody: Apoptosis is related to many diseases and development. The p53 tumor-suppressor protein induces apoptosis through transcriptional activation of several genes including p53R2, p53AIP1, and PUMA. A new p53 target gene, Noxa, was recently identified, which encodes a protein belonging to the subfamily of BH3-only proapoptic proteins. Noxa and PUMA are both transcriptional targets of p53 and BH3-only proteins. X-ray irradiation increased p53-dependent Noxa mRNA and protein levels. Noxa, when ectopically expressed, interacted with anti-apoptotic Bcl-2 family members, resulting in the activation of caspase-9. Noxa, like PUMA, localized to mitochondria and induces apoptosis in response to p53. Noxa and PUMA may represent direct mediators of p53-induced apoptosis. Increased levels of p53 and its target gene Noxa was found in the impaired tumor development.

SLAMF7 is a single-pass type I membrane protein and contains 1 Ig-like C2-type (immunoglobulin-like) domain. SLAMF7 is expressed in NK cells, activated B-cells, NK-cell line but not in promyelocytic, B-cell lines, or T-cell lines. Although the cytoplasmic domain of CS1 contains immunoreceptor tyrosine-based switch motifs (ITSM), which enables to recruite signaling lymphocyte activation molecule (SLAM)-associated protein (SAP/SH2D1A), it activates NK cells in the absence of a functional SAP. SLAMF7 positively regulated natural killer cell functions by a mechanism dependent on the adaptor EAT-2 but not the related adaptor SAP. However, in the absence of EAT-2, CRACC potently inhibited natural killer cell function. It was also inhibitory in T cells, which are typically devoid of EAT-2. Thus, SLAMF7 can exert activating or inhibitory influences on cells of the immune system depending on cellular context and the availability of effector proteins.
