There are several ways of labeling an oligonucleotide with fluorescein. The choice of label is diversified further, depending on the spectral requirements. 5'-fluorescein-CE phosphoramidite, derived from the single isomer 6-carboxyfluorescein, 5'-hexachlorofluorescein-CE phosphoramidite (HEX) and 5'-tetrachlorofluorescein-CE phosphoramidite (TET) can all be used to efficiently label an oligonucleotide at the 5'-end. Labeling the 3'-end of an oligo with fluorescein can be achieved using 3'-Fluorescein CPGs. Standard cleavage and deprotection with ammonium hydroxide liberates the fluorescein-labeled oligo when using any of these supports.

Dissolve the peptide in distilled, oxygen free water (oxygen free water is recommended to help prevent oxidation of the Cys groups). The concentration should be high enough to allow for addition of buffer after the product is dissolved completely. If necessary, a few drops of dilute acetic acid or ammonium hydroxide can be added to help solubilize the peptide. Brief sonication can also help in solubilization. Buffer or saline should be added only after the peptide is fully in solution. Solutions should be kept slightly basic to avoid degradation. Long term storage of solutions is not recommended.

A vitamin/enzyme cofactor found in milk, eggs, malted barley, liver, kidney, heart, leafy vegetables. Richest natural source is yeast. Occurs in the free form only in the retina of the eye, in whey, and in urine; bioactive forms occurring in tissues and cells are riboflavin monophosphate and flavine-adenine dinucleotide. Used as a photoinitiator for polymerization of polyacrylamide gels. It forms free radicals in aqueous solution in the presence of light. Riboflavin photodecomposes to leucoflavin. No free radicals are formed in the absence of oxygen but traces of oxygen allows for leucoflavin to reoxidize with free-radical generation. The catalysts TEMED or DMAPN are commonly added to speed up the free radical formation. The free radicals will cause acrylamide and bis-acrylamide to polymerize to form a gel matrix which can be used for electrophoresis. Riboflavin is commonly used in the stacking gel for non-denaturing polyacrylamide electrophoresis because native proteins can be senstive to persulfate ions from ammonium persulfate. Another advantage of riboflavin over ammonium persulfate is that it will not start polymerization until the gel is illuminated. Riboflavin causes less denaturing than other initiators. Gel strength can be improved when used with PTO (Pyrithione). Soluble in 0.1 M Sodium Hydroxide (50 mg/ml - clear, yellowish-orange solution); only slightly soluble in water.

D-Biotin is a growth factor present in small amounts in every living cell. It is involved in naturally occurring carboxylation reactions. It occurs mainly bound to proteins or polypeptides. It is more abundant in the liver, kidney, pancreas, yeast and milk. Biotin levels are higher in cancerous tumors than in normal tissues. It is inactivated by binding to avidin.
D-Biotin may be used to elute proteins from avidin/streptavidin resins. It is widely used for dietary supplements and fortified foods. It is also used for tablets and hard-shell capsule preparation due to its pharmaceutical properties.
Essential vitamin that is important for amino acid and energy metabolism, and fatty acid synthesis. It is a prosthetic group in four mammalian carboxylase families and facilitates the binding and transfer of carbon dioxide.
Soluble in water (22 mg/100 mL), ethanol (80 mg/100 mL), more soluble in hot water and in dilute alkalies; insoluble in other common organic solvents. Soluble in 2 M Ammonium hydroxide (50 mg/mL - clear, colorless solution), dimethylformamide (1.7 mg/mL).
