CD279 (Programmed Cell Death Protein 1; PD-1) is a type I transmembrane protein belonging to the CD28/CTLA-4 family of immunoreceptors that mediate signals for regulating immune responses. Members of the CD28/CTLA-4 family have been shown to either promote T cell activation (CD28 and ICOS) or downregulate T cell activation (CTLA-4 and PD-1). CD279 is expressed on activated T cells, B cells, myeloid cells and on a subset of thymocytes. In vitro, ligation of CD279 inhibits TCR-mediated T cell proliferation and production of IL-1, IL-4, IL-10 and IFN-gamma. In addition, CD279 ligation also inhibits BCR mediated signaling. Interleukin-4 (IL-4) is a cytokine produced by type 2 helper T cells, the Th2 cells. These cells tends to make a specific set of lymphokines including IL-4, IL-5, IL-6, IL-10, IL-13, IL-3 and GM-CSF and fail to produce IL-2, IFN-gamma, and lymphotoxin (TNF-beta). In addition, mast cells can produce IL-4. IL-4 exerts numerous effects on various hematopoietic cell types. On B cells, IL-4 promotes immunoglobulin class switching to IgE and IgG1 isotypes and upregulates MHC class II and CD23 expression. CX3CL1 is expressed on TNF-alpha and IL-1 activated endothelial cells (ECs) and binds to CX3CR1. CX3CL1 can exist in 2 forms, as membrane-anchored or as a shed 95-kDa glycoprotein. The soluble CX3CL1 has potent chemoattractant activity for T cells and monocytes, and the cell surface bound protein, which is induced on activated primary endothelial cells, promotes strong adhesion of those leukocytes. CX3CL1 may play a role in regulating leukocyte adhesion and migration processes at the endothelium.

CD278, also called inducible costimulator (ICOS) or CRP1 (CD28-related protein-1), is a member of the growing CD28 family of immune costimulatory receptors. Other family members are CD28, CTLA-4 and PD-1. ICOS is expressed on most CD45RO+ cells. ICOS expression is upregulated within approximately 24-48 hours of activation on Th primed cells. B7-H2, a member of the B7 family of costimulatory ligands, has been identified as the ICOS ligand. The B7-H2/ICOS interaction appears to play roles in T cell dependent B cell activation and Th differentiation.

Neuregulin-4 (Nrg4) belongs to a small family of EGF-like (EGFL) domain-containing proteins that are synthesized as transmembrane precursors and undergo proteolytic cleavage. The EGF-like domain (aa 5-46) of Nrg4 (aa 1-53) directly binds to the receptors ErbB3 and 4. Nrg4 is a cold induced adipokine, highly expressed in adipose tissues and enriched in brown fat. It is increased during brown adipocyte differentiation and reduced in rodent and human obesity. It promotes neurite outgrowth and protects against diet-induced insulin resistance and hepatic steatosis through attenuating hepatic lipogenic signaling. Inflammatory caspases (also known as group I caspases) are encoded by three main genes in humans, caspase-1, caspase-4 and caspase-5 and three main genes in mouse, caspase-1, caspase-11 and caspase-12. Caspase-11 is the murine orthologue of human caspase-4 and -5. Murine caspase-11 is a poorly characterized member of the caspase-1 subfamily.
