AIM2 is a strong candidate as a tumor suppressor as it is a member of the HIN-200 (hemopoietic interferon-inducible, nuclear proteins containing a 200 amino acid repeat) family of proteins that have been shown to regulate cell growth and survival. The HIN-200 family of proteins consists of four members in human, IFI 16, MNDA, AIM2 and IFIXl. Recently it has been reported that AIM2 is a receptor for cytosolic dsDNA, which forms a novel inflammasome complex with ASC to activate caspase-1-mediated processing of IL-1beta.

Podoplanin is expressed on glomerular epithelial cells (podocytes), type I lung alveolar cells, lymphatic endothelial cells and numerous tumors, including colorectal tumors, squamous cell carcinomas, testicular seminoma and brain tumors. Podoplanin is the ligand for C-type lectin-like receptor 2 (CLEC-2). Their association is dependent on sialic acid on O-glycans of podoplanin. Through its association with CLEC-2, podoplanin induces platelet aggregation and tumor metastasis. Podoplanin is also necessary for lymphatic vessel formation, normal lung cell proliferation and alveolus formation at birth.

N-Hexadecanoyl-DL-homoserine lactone (C16-HSL) is a small diffusible signaling molecule and is a member of N-acyl-homoserine lactone family. N-acylhomoserine lactones (AHL) are involved in quorum sensing, controlling gene expression, and cellular metabolism. C16-HSL is a lipophilic, long acyl side-chain bearing AHL, produced by the LuxI AHL synthase homolog SinI involved in quorum-sensing signaling in S. meliloti, a nitrogen-fixing bacterial symbiont of certain legumes. C16-HSL is the most abundant AHL produced by the proteobacterium R. capsulatus and activates genetic exchange between R. capsulatus cells. It tends to localize in relatively lipophilic cellular environments of bacteria and cannot diffuse freely through the cell membrane. The long-chain N-acylhomoserine lactones may be exported from cells by efflux pumps or may be transported between communicating cells by extracellular outer membrane vesicles.
