The activating transcription factor ATF-2 (also called CRE-BP1) binds to both AP-1 and CRE DNA response elements and is a member of the ATF/CREB family of leucine zipper proteins. ATF-2 has been implicated in the transcriptional regulation of a number of genes including cytokines, cell cycle control and apoptosis. Various forms of cellular stress, including inflammatory cytokines and UV irradiation, stimulate the transcriptional activity of ATF-2. Stress induced ATF-dependent transcription is dependent on phosphorylation of ATF. Serine 490 and serine 498 are novel phosphorylation sites on ATF that have recently been identified. In addition, ATF2 is particularly abundant in the brain and the ATF2 family of transcription factors is considered important substrates of signals upstream of the activation of genes associated with neuronal growth and differentiation. ATF expression has also been linked to the depression in humans.

Tyrosine hydroxylase is an excellent marker for dopaminergic and noradrenergic neurons. Tyrosine hydroxylase (a.k.a. tyrosine 3-monooxygenase) is the enzyme responsible for catalyzing the conversion of the amino acid L-tyrosine to L-3,4-dihydroxyphenylalanine (L-DOPA). L-DOPA is a precursor for dopamine, which, in turn, is a precursor for the important neurotransmitters norepinephrine (noradrenaline) and epinephrine (adrenaline). Tyrosine hydroxylase catalyzes the rate limiting step in this synthesis of catecholamines. In humans, tyrosine hydroxylase is encoded by the TH gene, and the enzyme is present in the central nervous system (CNS), peripheral symphatic neurons and the adrenal medulla. The enzymatic activity of TH requires ferrous ions as cofactors and is believed to be regulated by phosphorylation. At least four isoforms of human TH have been identified which result from alternative splicing. It stimulates migration of monocytes and T-lymphocytes through its receptors, CXCR4 and ACKR3, and decreases monocyte adherence to surfaces coated with ICAM-1, a ligand for beta-2 integrins. SDF1A/CXCR4 signaling axis inhibits beta-2 integrin LFA-1 mediated adhesion of monocytes to ICAM-1 through LYN kinase. It also plays a protective role after myocardial infarction, induces down-regulation and internalization of ACKR3 expressed in various cells and stimulates the proliferation of bone marrow-derived b progenitor cells in the presence of IL-7 as well as growth of the stromal cell-dependent B-cell clone DW34 cells.

The serum amyloid A family comprises a number of differentially expressed apolipoproteins, acute-phase SAA1 and SAA2, the former being the major component in plasma and constitutive SAAs. Although the liver is the primary site of synthesis of both SAA types extrahepatic production has been reported. The in vivo concentrations increase by as much as 1000 fold during inflammation. Several studies have expressed its importance in the diagnosis and monitoring of various diseases. Pathological SAA values are often detected in association with normal CRP concentrations; SAA rises earlier and more sharply than CRP. Recently, a broader view of SAA expression and function has been emerging. Expression studies show production of SAA proteins in histologically normal, atherosclerotic, Alzheimer, inflammatory, and tumour tissues. SAA has been found to have binding sites for high density lipoproteins, calcium, laminin, and heparin/heparin sulphate. Also adhesion motifs were identified and new functions affecting cell adhesion, migration, proliferation, and aggregation were discovered. These findings emphasize the importance of SAA in various physiological and pathological processes including inflammation, atherosclerosis, thrombosis, AA-amyloidosis, rheumatoid arthritis, and neoplasia. SAA has also a number of immunomodulatory roles, it can induce chemotaxis and adhesion molecule expression, has cytokine-like properties and can promote the upregulation of metalloproteinases. It enhances the binding of high density lipoprotein to macrophages and thus helps in the delivery of lipids to sites of injury for use in tissue repair, it is thus thought to be an integral part of the disease process.

Chromosomal passenger proteins are members of a group of proteins that move from centromeres to the spindle midzone during mitosis. INCENP (Inner Centromere Protein) is a chromosomal passenger protein with an essential role in mitosis and meiosis. It is a complex highly basic multidomain protein that is dynamically translocated from metaphase chromosomes to the spindle midzone during or just prior to anaphase. Some INCENP transfers to the equatorial cortex before formation of the cleavage furrow. Ultimately, it is discarded in the midbody at the completion of cytokinesis. INCENP, in complex with the passenger proteins aurora B kinase and survivin, acts at multiple points during mitosis. Studies in knockout mice established INCENP as essential for mouse development and viability. Loss of INCENP results in chromosome segregation defects and failures in cytokinesis. Targeting of survivin to its centromeric and central spindle locations during mitosis is dependent on INCENP. Direct binding of INCENP to beta tubulin has been demonstrated. Interactions of INCENP with heterochromatin protein HP1Hsa alpha and histone H2A.Z were also reported. INCENP levels are increased in several human cancer lines.
