Matrix metalloproteinases are members of a unique family of proteolytic enzymes that have a zinc ion at their active sites and can degrade collagens, elastin and other components of the extracellular matrix (ECM). However, overexpression and activation of MMPs have been linked with a range of pathological processes and disease states involved in the breakdown and remodeling of the ECM. Such diseases include tumor invasion and metastasis, rheumatoid arthritis, periodontal disease and vascular processes such as angiogenesis, intimal hyperplasia, atherosclerosis and aneurysms. Recently, MMPs have been linked to neurodegenerative diseases such as Alzheimer′s, and amyotrophic lateral sclerosis (ALS). Natural inhibitors of MMPs, tissue inhibitor of matrix metalloproteinases (TIMPs) exist and synthetic inhibitors have been developed which offer hope of new treatment options for these diseases.

Proinsulin C-peptide is the sequence of C peptide as it exists within proinsulin, the precursor to insulin which consists of insulin A chain, insulin B chain and C peptide (connecting peptide). In proinsulin, C peptide provides a means to ensure correct folding and assembly of the A and B chains. It is eventually cleaved away by proteases PC2, PC1/3 and CPE, at the flanking Arg-Arg and Lys-Arg basic residues (Arg-Arg-C peptide-Lys-Arg). Although C peptide is not present in mature insulin, it is stored in secretory granules, and eventually released into the bloodstream together with insulin in nearly equimolar amounts. Whereas insulin is metabolized quickly from circulation, C-peptide exhibits a slow turnover rate (>30 minutes). The measurement of the C-peptide is an important test for the β-cell function. In the red blood cells of type 2 diabetic patients, Na+,K+, ATPase activity is strongly related to blood C-peptide levels. C-peptide signal transduction in human renal tubular cells involves the activation of phospholipase C and PKC-δ and PKC-varepsilon, as well as RhoA, followed by phosphorylation of ERK1/2 and JNK and a parallel activation of Akt. C-peptide shows specific binding to a G-protein-coupled membrane binding site, resulting in Ca2+ influx, activation of mitogen-activated protein kinase signalling pathways and stimulation of Na+, K+ ATPase and endothelial nitric oxide synthase.

Chromosomal passenger proteins are members of a group of proteins that move from centromeres to the spindle midzone during mitosis. INCENP (Inner Centromere Protein) is a chromosomal passenger protein with an essential role in mitosis and meiosis. It is a complex highly basic multidomain protein that is dynamically translocated from metaphase chromosomes to the spindle midzone during or just prior to anaphase. Some INCENP transfers to the equatorial cortex before formation of the cleavage furrow. Ultimately, it is discarded in the midbody at the completion of cytokinesis. INCENP, in complex with the passenger proteins aurora B kinase and survivin, acts at multiple points during mitosis. Studies in knockout mice established INCENP as essential for mouse development and viability. Loss of INCENP results in chromosome segregation defects and failures in cytokinesis. Targeting of survivin to its centromeric and central spindle locations during mitosis is dependent on INCENP. Direct binding of INCENP to beta tubulin has been demonstrated. Interactions of INCENP with heterochromatin protein HP1Hsa alpha and histone H2A.Z were also reported. INCENP levels are increased in several human cancer lines.
