Gamma-aminobutyric acid (GABA) is the primary inhibitory neurotransmitter in the central nervous system, causing a hyperpolarization of the membrane through the opening of a Cl- channel associated with the GABAA-Receptor (GABAA-R) subtype. GABAA-Rs are important therapeutic targets for a range of sedative, anxiolytic, and hypnotic agents and are implicated in several diseases including epilepsy, anxiety, depression, and substance abuse. The GABAA-R is a multimeric subunit complex. To date six alphas, four betas and four gammas, plus alternative splicing variants of some of these subunits, have been identified. Injection in oocytes or mammalian cell lines of cRNA coding for alpha and beta subunits results in the expression of functional GABAA-Rs sensitive to GABA. However, coexpression of a gamma subunit is required for benzodiazepine modulation. The various effects of the benzodiazepines in brain may also be mediated via different alpha subunits of the receptor. Lastly, phosphorylation of beta subunits of the receptor has been shown to modulate GABAA-R function.

PTEN indirectly activates the AKT/PI3K pathway, an important signaling pathway for cell growth and proliferation by keeping levels of the second messenger PIP3 low, thereby preventing phosphoinositide-dependent kinase-1 (PDK-1) from phosphorylating and activating AKT. Expression of wild-type PTEN causes growth arrest in many cancer cell lines, but expression of a PTEN protein containing a mutation that blocks its lipid phosphatase activity failed to suppress growth of glioma cell lines suggesting that the tumor suppressive effect of PTEN is mediated solely by its lipid phosphatase activity. Other activities include the inhibition of insulin stimulated MAPK activation by blocking the insulin-receptor substrate (IRS)-1 phosphorylation and assembly of the IRS-1/Grb2/Sos complex.

Human Interleukin family 1 Member 7 (IL1F7) is a member of the Interleukin 1 cytokine family. Five alternatively spliced transcript variants encoding distinct isoforms have been reported with distinct expression profiles. The longest IL1F7 transcript, referred to as IL1F7b or IL1F7 isoform 1, encodes a 218 amino acid residues proprotein containing a 45 amino acid propeptide, which is cleaved to generate mature protein. IL1F7b binds to IL18 R alpha with low affinity but does not exert any IL18 agonistic or antagonistic effects. IL1F7b also binds interleukin 18 binding protein (IL-18BP), an inhibitory binding protein of interleukin 18 (IL-18), and subsequently forms a complex with IL18 receptor beta subunit, and through which it inhibits the activity of IL-18.
