Human Recombinant Galectin-3 (from E. coli)
Supplier: Peprotech
Lectins, of either plant or animal origin, are carbohydrate-binding proteins that interact with glycoproteins and glycolipids on the surface of animal cells. The Galectins are lectins that recognize and interact with β-galactoside moieties. Galectin-3 regulates a number of biological processes, including embryogenesis, inflammatory responses, cell progression and metastasis. Galectin-3 is normally expressed in epithelia of a variety of tissues, including colon and endometrium, and in various inflammatory cells, including macrophages. Galectin-3 can function intracellularly, controlling the cell cycle and preventing T-cell apoptosis, and also extracellularly, by activating various cells, including monocytes/macrophages, mast cells, neutrophils, and lymphocytes. Expression of Galectin-3 is affected by neoplastic transformation, being up-regulated in certain types of lymphomas, and in thyroid and hepatic carcinomas. Conversely, it is down-regulated in other cancers such as colon, breast, ovarian, and uterine. Recombinant Human Galectin-3 is a globular 26.0 kDa protein containing 250 amino acid residues, but no disulfide bonds.

Anti-SLC35D1 Rabbit Polyclonal Antibody
Supplier: Rockland Immunochemical
The solute carrier family Slc35 consists of at least 17 proteins that act as nucleotide sugar transporters localized to the Golgi apparatus and endoplasmic reticulum. The role of the ER-resident Slc family member Slc35D1 is to transport both UDP-glucuronic acid and UDP-N-acetylgalactosamine. These molecules can serve as substrates for chondroitin sulfate biosynthesis and mice lacking the Slc35D1 gene developed a lethal form of skeletal dysplasia with severe shortening of limbs and facial structures. Examination of epiphyseal cartilage in these mice revealed a decreased proliferating zone with round chrondrocytes, scarce matrices, and reduced proteoglycan aggregates. Loss of function mutations in human Slc35D1 cause Schneckenbecken dysplasia, a severe skeletal dysplasia.

Anti-MSI2 Rabbit Polyclonal Antibody
Supplier: Rockland Immunochemical
Musashi2 (MSI2) is an RNA-binding protein that is highly expressed in precursor cells in the ventricular and subventricular zones of the developing mammalian CNS. Like the related MSI1, MSI2 has been suggested to be involved stem cell production and maintenance. MSI2 is the predominant MSI protein in hematopoietic stem cells, and its knockdown leads to reduced engraftment and depletion in vivo. Expression levels of MSI2 are elevated in myeloid leukemia cells lines, and MSI2 appears to cooperate with BCR-ABL1 to induce an aggressive leukemia; the level of MSI2 directly correlates with decreased survival in patients. MSI2 negatively regulates the asymmetric cell fate determinant NUMB, suggesting that this signaling pathway may provide future targets for future therapies.

Anti-NAA35 Rabbit Polyclonal Antibody
Supplier: Prosci
MAK10 Antibody: The MAK10 gene encodes a 733-amino acid protein with several regions of similarity to T cell receptor alpha-subunit V (variable) regions in yeast. The mammalian homologue of yeast MAK10, also known as EGAP, is one subunit of a novel N-terminal acetyltransferase (NAT) that is highly conserved among vertebrate species. It is expressed in a variety of tissues in the developing rat embryo but restricted in expression in the adult, remaining detectable only in tissues undergoing continual cell renewal or in cells responding to pathological injury. The MAK10-NAT complex is an essential regulatory enzyme controlling the function of a subset of proteins required for embryonic growth control and vessel development. This complex functionally co-assembles in mammalian cells to regulate cell proliferation and is essential for embryonic development, at least in part through the regulation of target of rapamycin (TOR) signaling events. At least two isoforms of MAK10 are known to exist.

Human Recombinant IL-12 p70 (from HEK293 cells)
Supplier: Peprotech
IL-12 is a potent regulator of cell-mediated immune responses and it induces IFN-gamma production by NK and T cells. It is produced by activated monocytes/macrophage cells, B lymphocytes and connective tissue-type mast cells. Among its biological activities, IL-12 promotes the growth and activity of activated NK, CD4+ and CD8+ cells, and induces the development of IFN-gamma-producing Th1 cells. PeproTech's Recombinant Human IL-12 p70, derived from HEK293 cells, is a heterodimeric glycoprotein consisting of disulfide-linked p35 and p40 subunits (503 total amino acid residues). The calculated molecular weight of Human IL-12 p70 is 57.2 kDa; however, due to glycosylation, it migrates at an apparent molecular weight of 65-70 kDa based on SDS-PAGE gel, under non-reducing conditions.

Anti-XAF1 Rabbit Polyclonal Antibody
Supplier: Rockland Immunochemical
XAF-1 binds to XIAP, an inhibitor of caspases-3, -7, and -9, and triggers its relocation from the cytosol to the nucleus. Overexpression of XAF-1 results in the neutralization of XIAP's ability to inhibit cell death. XAF-1 is normally expressed in all adult and fetal tissues but was found to be present in very low levels in a variety of cancer cell lines. In contrast, XIAP levels have been shown to be high in a majority of cell lines. Low XAF-1 and high basal expression of XIAP may therefore play a critical role in maintaining survival of cancer cell lines. Both IFN-alpha2 and IFN-beta can induce XAF-1 mRNA in all cells examined but induction of XAF-1 protein (as observed by immunoblot analysis) was seen only in cell lines sensitive to the apoptotic effects of IFNs.

Acrylamide ≥99.9%, white crystalline powder
Supplier: MP Biomedicals
Acrylamide is a monomer used in a variety of synthetic processes to form polymers and copolymers. It polymerizes in the presence of free radicals in aqueous solutions. It is usually used to prepare polyacrylamide gels for electrophoresis separation of biomolecules. Common reaction initiators are riboflavin and ammonium persulfate; varying the ratio of acrylamide to crosslinking agent permits the formation of a gel with predictable average pore size and texture. Acrylamide tends to hydrolyze under acidic or basic conditions to form acrylic acid. Polyacrylamide plastics have numerous commercial applications, due in part to the tendency of polyacrylamide to adsorb many times its own mass in water. Acrylic acid and any ionic impurities in the acrylamide can have significant effects on the performance of the acrylamide gel formed, since the voltage across the gel is affected by the ionic charge of the gel and usage buffers.
Store at Room Temperature (15-30 °C), Dessicated.

Anti-TNFSF11 Rabbit Polyclonal Antibody
Supplier: Rockland Immunochemical
The receptor activator of NF-kB ligand (RANK-L) is a recently discovered member of the TNF-ligand family involved in the regulation of the T cell-dependent immune response, lymph node organogenesis and bone formation. RANK-L exists as both a normal, transmembrane form and a truncated, soluble form (sRANK-L), both of which can stimulate the receptor. Activation of T cells, such as by treatment with interleukin-7, induces RANK-L production and leads to an increase of osteoclast formation and bone loss. Finally, sRANK-L can activate the antiapoptotic kinase Akt through a signaling complex involving Src kinase and TRAF6, suggesting sRANK-L may also play a role in regulating apoptosis. This antibody will recognize both the soluble form and the uncleaved transmembrane form of RANK-L.

Sulfo-SMA Membrane protein solubilization & stabilization
Supplier: CUBE BIOTECH
Sulfo-SMA is an electroneutral modification of existing SMAs. It does not interfere with charge-sensitive interactions between proteins and lipids. This innovation opens up a wider range of experimental research in terms of charge-sensitive membrane protein processes like protein-protein and protein-lipid interactions. In addition, Sulfo-SMA belongs to a new generation of SMA’s which are RAFT polymerized. This achieves a reduction in both monomer size and greater monodispersity.
Another significant advantage of Sulfo polymers compared to other polymers is the wide pH range in which they remain stable. The buffer in which the polymer is supplied has a pH of 7.5, but the polymer itself remains stable between pH 4 and pH 10.
The special physicochemical properties of Sulfo-SMAs make them ideal for cryo-TEM and other downstream applications.
After successful membrane protein solubilization, the protein can be purified using affinity chromatography. For membrane protein purification, we recommend using the Rho1D4-tag. Cube Biotech offers matching products for this purpose.
