The solute carrier family Slc35 consists of at least 17 proteins that act as nucleotide sugar transporters localized to the Golgi apparatus and endoplasmic reticulum. The role of the ER-resident Slc family member Slc35D1 is to transport both UDP-glucuronic acid and UDP-N-acetylgalactosamine. These molecules can serve as substrates for chondroitin sulfate biosynthesis and mice lacking the Slc35D1 gene developed a lethal form of skeletal dysplasia with severe shortening of limbs and facial structures. Examination of epiphyseal cartilage in these mice revealed a decreased proliferating zone with round chrondrocytes, scarce matrices, and reduced proteoglycan aggregates. Loss of function mutations in human Slc35D1 cause Schneckenbecken dysplasia, a severe skeletal dysplasia.

Lectins, of either plant or animal origin, are carbohydrate-binding proteins that interact with glycoproteins and glycolipids on the surface of animal cells. The Galectins are lectins that recognize and interact with β-galactoside moieties. Galectin-3 regulates a number of biological processes, including embryogenesis, inflammatory responses, cell progression and metastasis. Galectin-3 is normally expressed in epithelia of a variety of tissues, including colon and endometrium, and in various inflammatory cells, including macrophages. Galectin-3 can function intracellularly, controlling the cell cycle and preventing T-cell apoptosis, and also extracellularly, by activating various cells, including monocytes/macrophages, mast cells, neutrophils, and lymphocytes. Expression of Galectin-3 is affected by neoplastic transformation, being up-regulated in certain types of lymphomas, and in thyroid and hepatic carcinomas. Conversely, it is down-regulated in other cancers such as colon, breast, ovarian, and uterine. Recombinant Human Galectin-3 is a globular 26.0 kDa protein containing 250 amino acid residues, but no disulfide bonds.

Acrylamide is a monomer used in a variety of synthetic processes to form polymers and copolymers. It polymerizes in the presence of free radicals in aqueous solutions. It is usually used to prepare polyacrylamide gels for electrophoresis separation of biomolecules. Common reaction initiators are riboflavin and ammonium persulfate; varying the ratio of acrylamide to crosslinking agent permits the formation of a gel with predictable average pore size and texture. Acrylamide tends to hydrolyze under acidic or basic conditions to form acrylic acid. Polyacrylamide plastics have numerous commercial applications, due in part to the tendency of polyacrylamide to adsorb many times its own mass in water. Acrylic acid and any ionic impurities in the acrylamide can have significant effects on the performance of the acrylamide gel formed, since the voltage across the gel is affected by the ionic charge of the gel and usage buffers.

Another significant advantage of Sulfo polymers compared to other polymers is the wide pH range in which they remain stable. The buffer in which the polymer is supplied has a pH of 7.5, but the polymer itself remains stable between pH 4 and pH 10.
The special physicochemical properties of Sulfo-SMAs make them ideal for cryo-TEM and other downstream applications.
After successful membrane protein solubilization, the protein can be purified using affinity chromatography. For membrane protein purification, we recommend using the Rho1D4-tag. Cube Biotech offers matching products for this purpose.

PEDF is a noninhibitory serpin with neurotrophic, anti-angiogenic, and anti-tumorigenic properties. It is a 50 kDa glycoprotein produced and secreted in many tissues throughout the body. A major component of the anti-angiogenic action of PEDF is the induction of apoptosis in proliferating endothelial cells. In addition, PEDF is able to inhibit the activity of angiogenic factors such as VEGF and FGF-2. The neuroprotective effects of PEDF are achieved through suppression of neuronal apoptosis induced by peroxide, glutamate, or other neurotoxins. The recent identification of a lipase-linked cell membrane receptor for PEDF (PEDF-R) that binds to PEDF with high affinity (1) should facilitate further elucidation of the underlying mechanisms of this pluripotent serpin. To date, PEDF-R is the only signaling receptor known to be used by a serpin family member. The unique range of PEDF activities implicate it as a potential therapeutic agent for the treatment of vasculature related neurodegenerative diseases such as age-related macular degeneration (AMD) and proliferative diabetic retinopathy (PDR). PEDF also has the potential to be useful in the treatment of various angiogenesis-related diseases including a number of cancers.
