Polyspecific organic cation transporters in the liver, kidney, intestine, and other organs are critical for elimination of many endogenous small organic cations as well as a wide array of drugs and environmental toxins. The encoded protein is a plasma integral membrane protein which functions both as an organic cation transporter and as a sodium-dependent high affinity carnitine transporter. The encoded protein is involved in the active cellular uptake of carnitine. Mutations in this gene are the cause of systemic primary carnitine deficiency (CDSP), an autosomal recessive disorder manifested early in life by hypoketotic hypoglycemia and acute metabolic decompensation, and later in life by skeletal myopathy or cardiomyopathy. SPP24, also known as secreted phosphoprotein 2, is a 211 amino acid secreted protein that belongs to the cystatin superfamily. Expressed in liver and plasma, SPP24 may play a role in coordinating an aspect of bone turnover. The MDC gene is one of several Cys-Cys (CC) cytokine genes which are clustered on the q arm of chromosome 16. Cytokines are a family of secreted proteins involved in immunoregulatory and inflammatory processes. The CC cytokines are proteins characterized by two adjacent cysteines. The cytokine encoded by MDC displays chemotactic activity for natural killer cells, chronically activated T lymphocytes, monocytes and dendritic cells. It has no chemoattractant activity for eosinophils, neutrophils and resting T lymphocytes and also displays a mild activity for primary activated T lymphocytes. The product of this gene binds to chemokine receptor CCR4. This chemokine may play a role in the trafficking of activated T lymphocytes to inflammatory sites and other aspects of activated T lymphocyte physiology.

TMEM38A and TMEM38B are two recently identified trimeric intracellular cation (TRIC) channel subtypes. SHISA4 plays an essential role in the maturation of presomitic mesoderm cells by individual attenuation of both FGF and WNT signaling. The Shisa family of single-transmembrane proteins is characterized by an N-terminal cysteine-rich domain and a proline-rich C-terminal region. Its founding member, Xenopus Shisa, promotes head development by antagonizing Wnt and FGF signaling. Shisa physically interacted with immature forms of the Wnt receptor Frizzled and the FGF receptor within the ER and inhibited their posttranslational maturation and trafficking to the cell surface. Loss of Shisa function sensitized the neuroectoderm to Wnt signaling and suppressed head formation during gastrulation.

KLRA1 (also known as Ly49L) is a member of the LY49 family of receptors in Natural Killer (NK) cells that bind to major histocompatibility complex (MHC) class 1. MARCH8 (c-MIR) is a novel E3 ubiquitin ligase designated as the modulator of immune recognition (MIR) family, whose catalytic domain is a variant RING domain (RING-CH domain). MARCH8 was found as a functional and structural homolog of KSHV MIR1 and MIR2. MARCH8 targets B7-2 to lysosomal degradation and down-regulates the B7-2 surface expression through ubiquitination of its cytoplasmic tail. Furthermore, MARCH8 has been shown to down-regulate the expression of transferrin receptor and Fas, an important molecule for the induction of apoptosis. MARCH8 is the first example of an E3 ubiquitin ligase that is capable of inhibiting MHC II expression. ZNF346, also known as JAZ (just another zinc-finger protein), is a nucleolar, zinc finger protein which preferentially binds to double-stranded (ds) RNA or RNA/DNA hybrids with high affinity via C2H2 zinc fingers. ZNF346 contains four CH-type zinc finger motifs that are connected by long (28-38) amino acid linker sequences. ZNF346 is expressed in all tissues tested and localizes to the nucleus, primarily the nucleolus. ZNF346 is exported by exportin-5 but translocates back into nuclei by a facilitated diffusion mechanism. ZNF346 interacts with ILF3 in an RNA-independent manner and may be involved in cell growth and survival.
